Objective: To explore the relationship between the expression of CD47 molecule in patients with newly diagnosed adult acute myeloid leukemia(AML)and clinical prognosis,and to further explore the effect of azacitidine on the expression of CD47 molecules in HL-60 cell lines.Methods: Twenty cases with newly diagnosed AML in Our Hospital from April 2020 to October 2021 were analyzed,and five cases were regarded as the control group,including two cases with iron deficiency anemia,three cases with idiopathic immune thrombocytopenia.The expression of CD47 m RNA in mononuclear cells from bone marrow or peripheral blood was detected by real-time fluorescence quantitative polymerase chain reaction(q PCR).Relationship between the CD47 expression and demographic characteristics,risk stratification,complete remission and recurrence was observed.The overall survival(OS)was predicted by the Kaplan-Meier curve.The expression of CD47 m RNA was detected in HL-60 cell line in vitro by action of different concentrations of azacitidine.Results:(1).The expression of CD47 m RNA in mononuckear cells from bone marrow(P=0.0115)and peripheral blood(P=0.0069)was significantly higher in 20 patients with AML compared with 5 controls(2 iron deficiency anemia and 3 primary immune thrombocytopenia);(2).There was no correlation between the expression of CD47 m RNA in bone marrow cells in AML patients and complete blood counts(including leukocytes,hemoglobin,platelets),chromosome karyotypes,gene mutations,bone marrow immune expression of CD34,CD38 and CD123(P> 0.05);(3).There was no correlation betweenthe expression of CD47 m RNA of peripheral blood cells in AML patients and complete blood counts(including leukocytes,hemoglobin,platelets),primary cell percentage,gene mutations,chromosome karyotype,and clinical prognosis(P> 0.05);(4).The survival period from the high CD47 m RNA expression of bone marrow mononuclear cells(12 / 19 cases)was significantly decrease than that the low group(7 / 19 cases)(P=0.012);(5).The recurrence of AML included 7 cases with high expression(7 / 12)and 1 patient with low expression(1 / 7),indicating the increased risk of recurrence in patients with high expression(P=0.017);(6).The azacitidine half inhibition rate(IC50)of HL-60 cell line was 3.283 um / ml;the expression of HL-60 CD47 m RNA(0.4um,4um,16um)(P<0.05)with the same drug concentration(04 h,48h)(24h)(P <0.05).Conclusions:(1).The expression of CD47 m RNA increased in mononuclear cells from bone marrow and peripheral blood in adult patients with AML,but the expression of CD47 m RNA in bone marrow was more significant than peripheral blood expression.(2).There was no correlation between the expression of CD47 m RNA in complete blood counts(white blood cells,hemoglobin,platelets),percentage of primitive cells,gene mutation and chromosome karyotype(P>0.05);(3).High expression of CD47 m RNA in bone marrow mononuclear cells in adults with AML was associated with reduced survival and increased risk of recurrence,whereas high expression of CD47 m RNA in peripheral blood mononuclear cells in adults with primary AML was not significantly associated with survival and prognosis.(4).Azacitidine can downregulate the expression of CD47 m RNA in HL-60 cells.Figure (eleven) table (seven) reference (fifty-three)...
