Objective:To explore the expression and biological function of G protein-coupled receptor183 in laryngeal squamous cell carcinoma,and to evaluate the clinical significance of GPR183 in combination with patient clinical data,so as to provide a new theoretical basis for the clinical diagnosis and treatment of patients with laryngeal squamous cell carcinoma.Methods:Analyze the relevant data in the TCGA database to obtain the expression of GPR183 in LSCC,and the expression of GPR183 in LSCC and normal mucosal tissues were detected by immunohistochemistry and its clinical significance was analyzed by statistical analysis.The expression of GPR183 in the LSCC cell line was detected by q RT-PCR.After knocked down the expression level of GPR183 in LSCC cell lines using si RNA.To explore the function of GPR183 in laryngal squamous cell lines in vitro,CCK8 assay,clone formation,transwell and flow cytometry were used.Additionally,the xenograft tumor model in nude mice was established and the effect of GPR183 on the growth of LSCC was evaluated by intratumoral injection of si-GPR183 stable oligo.Finally,we use database to predict micro RNA targeting GPR183.Results:GPR183 was overexpressed in LSCC tissues and cells,which was associated with the malignant progression of LSCC.Silencing GPR183 expression level in LSCC cell lines can significantly inhibit the proliferation,migration and invasion of tumor cells,and induce cell cycle arrest at G1 phase,but has no statistically significant effects on the apoptosis of LSCC cells.In vivo experiments showed that GPR183 could significantly promote the growth of LSCC cells.The database predicts 5 micro RNAs,which has a post transcriptional regulatory effect on GPR183.Conclusion:The expression level of GPR183 in LSCC was significantly higher than that in adjacent mucosa,and its high expression was closely related to clinical T stage and lymph node metastasis of laryngeal squamous cell carcinoma.GPR183 is a potential oncogene in LSCC.Silencing GPR183 can not only significantly inhibit the proliferation of laryngeal squamous cell carcinoma cells,but also induce cell cycle arrest in G1 phase,and inhibit cell migration and invasion.Therefore,GPR183 may become a potential molecular target for the diagnosis,treatment and prognosis of LSCC.
