Abstract/Summary:
Diabetes is a common chronic disease that seriously harms human health. One of the most common and critical complications of diabetes is diabetic foot ulcer(DFU).DFU patients generally suffer from non-healing chronic wounds or even amputation at the later stage of the disease.Current researches mainly focus on effective treatment after the occurrence of DFU,but for diabetics,it may be more desirable to delay or prevent DFU before its development. Here,we developed a hierarchical microparticles(HMPs)-based microneedle(MN)patch, namely Z-H-MN-VEGF&b FGF(Zn OHMPs-MN-VEGF&b FGF),which could deliver drugs to the limbs painlessly, accurately, and controllably under near-infrared(NIR)irradiation.Therein the HMPs not only performed satisfactory photothermal ability, but also could effectively remove reactive oxygen species(ROS),thereby preventing its damage to blood vessels.Simultaneously, vascular endothelial growth factor and basic fibroblast growth factor(VEGF&b FGF)with good angiogenesis promoting ability and zinc oxide(Zn O)with excellent antibacterial activity were also introduced in this therapeutic platform. The synergistic effect of these materials encapsulated in the microneedle could not only combat excessive ROS and possible bacterial infections, but also promote angiogenesis before the development of DFU in diabetic rats. Therefore, this therapeutic platform is expected to become a painless and accurate method for the early prevention of DFU.
