Pueraria powder is a lump or granular material obtained by cleaning,peeling,crushing,filtering and drying the roots of pueraria liana of legumes.Pueraria powder in the effective components of the curative effect is accurate,a wide range of effects,food and health care industry consumers love.However,relevant articles have shown that pueraria root extract has certain hepatotoxicity and estrogen-like effects,and the preliminary experimental conclusion of our research group shows that it is relatively safe to consume 25 g/kg pueraria root water extract.Therefore,based on previous research conclusions,this study comprehensively and systematically evaluated the safety of Pueraria powder in SD rats by feeding Pueraria powder for90 days,followed by serum biochemical examination,histopathological examination and serum metabonomics analysis.The main results are as follows:1.Safety study of Pueraria powder in ratsIn the experiment of feeding Pueraria powder to the same group of normal rats,it was found that Pueraria powder had sex difference effect on male and female rats.Compared with blank control group,no abnormality was found in wet weight,organ coefficient,blood routine index and histopathological section between different dose groups in the experimental group.Dose in low dose group and male,female group of biochemical indicators of liver of rats found no obvious abnormality,the male high dose group of rats in biochemical indicators of liver AST,ALP,ALT,TBIL,DBIL showed significant differences,combined with the liver and liver biopsy results antioxidant index as a result,prompt the dose puerarin powder on male rats have effects on the liver,There was no instrumental injury.Pueraria root powder had a certain effect on the biochemical indexes of serum renal function of male and female rats,which showed that the Cr indexes of female low-dose and medium-dose groups and male low-dose groups were increased,but there was no significant difference in the high-dose group.UA index increased in female low-dose group and decreased significantly in male high-dose group,but there was no statistical significance in other dose groups.Combined with the renal organ coefficient,histopathological section and renal oxidation index of each group,all showed normal,suggesting that Pueraria powder did not cause substantial pathological changes in kidney.There was no significant difference in sex hormone index between male and female groups.Combined with organ coefficient and pathological section,Pueraria powder did not affect sex hormone secretion and sex organ development of rats.2.Safety study of Pueraria powder based on serum metabolomics in ratsThe metabolomics studies of the serum of the hermaphroditic drug group were carried out,and the main metabolic pathways involved in the differential metabolites screened were aminoacyl t RNABiosynthesis,cysteine and methionine metabolism,phenylalanine,winding and amino acid tryptophan biosynthesis,primary bile acid biosynthesis,glycerol phosphate metabolism,glycine,serine and threonine metabolism,glyceride metabolism,arginine biosynthesis,arginine and proline metabolism,pyrimidine metabolism,purine metabolism,tryptophan metabolism,glyoxylic acid,niacin,and nicotinic acid and dicarboxylic acid metabolism There were 16 metabolic pathways including amine metabolism,linoleic acid metabolism and histidine metabolism.Combined with the-log(p)>2 of pyrimidine metabolism in the male high-dose group and the pathway impact >0.1,the metabolic pathway was more disturbed.It was revealed that taking high doses of Pueraria powder for 90 consecutive days may have a certain effect on liver function in male rats,and the main manifestation of serum metabolism is a significant decrease in pyrimidine metabolism.Conclusion:From the detection of serum biochemical indicators and histopathological results of rats,although high dose(16.67 g/kg)of Pueraria powder had a certain effect on the AST,ALP,ALT,TBIL and DBIL indicators of male rat liver,liver slices did not show qualitative damage.Serum metabolomics studies have shown a significant decrease in pyrimidine metabolism,so Pueraria powder is a relatively safe medicinal and food dual-use substance.
