Objective:By establishing the exogenous model and endogenous-exogenous model of dampness-heat syndrome,explore the essence of dampness-heat syndrome,observe the changes in the expression of glucolipid metabolism,inflammatory factors and water-liquid metabolism and the influence of the intervention of Modified Wendan Decoction on the above indicators.Methods:Male SPF SD rats were randomly divided into 7 groups,including normal temperature control group,exogenous model control group,exogenous Xianglian Pill group,exogenous Modified Wendantang group,endogenous-exogenous model control group,endogenous-exogenous Xianglian Pill group,endogenous-exogenous Modified Wendantang group,with 10 rats in each group.The model of damp-heat syndrome was replicated by comparing the double effects of artificial climate box+ high-fat and high-sugar diet with the single effects of artificial climate box.After 10 days of modeling,the drug was administered by gavage for 4 weeks.The normal temperature control group and the model control group were given the same volume of water.After the last administration,the abdominal aortic blood,stomach,lung,kidney,liver and visceral fat tissues were collected by anesthesia.Visceral fat tissue was weighed and recorded for data analysis;Glucose(GLU)was detected by oxidase method;The serum triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were detected by microplate method;The tumor Necrosis Factor Alpha(TNF-α),Interleukin 1Beta(IL-1β),prostaglandin E2(PGE2),motilin(MTL),quaporins1~5(AQP1~5)in different tissues of rats were detected by enzyme-linked immunosorbent assay(ELISA);liver pathological sections was prepared by Paraffin section and HE staining.Results:1.The exogenous model and endogenous-exogenous model impact on the general condition,body weight and visceral fat ratio.Compared with the normal temperature control group,the general condition is poor,the performance is irritable,food intake is reduced,hair is dim into clusters with a small amount of shedding,soft stool and perianal fecal adhesion in the exogenous model control group and the endogenous-exogenous model control group.Compared with the normal temperature control group,the body weight and the visceral fat ratio in exogenous model control group significantly decreased(P<0.05 or P<0.01),the body weight in endogenous-exogenous model control group significantly decreased(P<0.05),the visceral fat ratio had no obvious decreasing trend.2.The exogenous model and endogenous-exogenous model impact on the glucolipid metabolism.Compared with the normal temperature control group,there was no significant difference in serum GLU,TG and TC content in the exogenous model control group and the endogenous-exogenous model control group,while LDL-C content significantly increased(P<0.05),HDL-C content was significantly decreased(P<0.05).The exogenous model and endogenous-exogenous model impact on the inflammatory factors and AQPs.Compared with the normal temperature control group,TNF-α in kidney and stomach,IL-1β in stomach,PGE2 in serum,AQP2 in kidney,AQP4 in stomach of the exogenous model control group were significantly increased(P<0.05 or P<0.01),TNF-α in kidney and stomach,IL-1β in kidney,PGE2 in serum,AQP5 in lung,AQP2 and AQP3 in kidney,AQP3 and AQP4 in stomach of the endogenous-exogenous model control group were significantly increased(P<0.05 or P<0.01).4.Modified Wendantang decoction can improve the general condition,reduce body weight and adjust glucolipid metabolism disorder.After medication was administered of the Modified Wendantang group,the general condition of rats was improved.The spirit of rats improved and their fighting reduced,the color of the stool turned from yellow into dark brown,and the stool formation was damper,the crissal adhesion reduced.Supplemented Modified Wendantang can reduce the body weight and visceral fat rate of rats in the exogenous model control group and the endogenous-exogenous model control group(P<0.05 or P<0.01),and the effect was most obvious in the endogenous-exogenous model control group.The contents of GLU,TG,TC,LDL-C were significantly decreased and the HDL-C was significantly increased(P<0.05 or P<0.01).5.Modified Wendantang decoction can reduce the content of inflammatory factors,improve the abnormal metabolism of water and liver cell damage.Modified Wendantang decoction can reduce the content of TNF-α in kidney and stomach,IL-1β and MTL in stomach,AQP1 in lung,AQP2 and AQP3 in kidney,AQP3 and AQP4 in stomach by the exogenous model control group(P<0.05 or P<0.01),TNF-αin kidney and stomach,IL-1β in kidney,PGE2 in serum,MTL in stomach,AQP1 in lung,AQP2 and AQP3 in kidney,AQP3 in stomach by the endogenous-exogenous model control group(P<0.05 or P<0.01).Modified Wendantang decoction can improve the steatosis and inflammatory infiltration of hepatocytes.Conclusion:1.Modified Wendantang decoction can reduce body weight and visceral fat ratio in the exogenous damp-heat model and the endogenous-exogenous damp-heat model,the effect was most obvious in the endogenous-exogenous model control group.2.Modified Wendantang decoction had a good callback effect on the glucolipid metabolism disorder of the exogenous model control group and the endogenous-exogenous model control group,there was no significant difference between the two models.3.Modified Wendan Decoction can reduce the content of the inflammatory factors in the external and internal damp-heat model and the internal and external damp-heat model,improve the abnormal water metabolism and liver cell damage,and the effect is most obvious in the endogenous-exogenous model control group.4.The molecular mechanism of modified Wendantang Decoction in the treatment of damp-heat syndrome is mainly related to lowering blood glucose,regulating blood lipids,down-regulating inflammatory factors and various AQPs,and improving liver cell injury,and the effect is most obvious in the endogenous-exogenous damp-heat syndrome. |
