| Objective:Taking Osteopontin(OPN)as the research target,the analysis of interleukin-25(Interleukin-25,IL-25),interleukin-33(Interleukin-33,IL-33),thymic stromal lymphocytes in a mouse model of bronchial asthma The effects of Thymic stromal lymphopoietin(TSLP)on OPN were evaluated,and the roles and correlations of OPN and IL-25,IL-33 and TSLP in airway remodeling were evaluated.Methods:1.Mice were randomly divided into 5 groups,divided into normal group,asthma model group,anti-IL-25 neutralizing antibody group,anti-IL-33 neutralizing antibody group,and anti-TSLP neutralizing antibody group.After the classical mouse asthma model was established by intraperitoneal injection and aerosol challenge of OVA,the expressions of OPN,IL-25,IL-33 and TSLP in the lung tissue of mice were detected by immunohistochemistry,Western Blot and RT-PCR.The expression of OPN,IL-25,IL-33 and TSLP in BALF and serum;2.The mice in the experimental group were affected by anti-IL-25 neutralizing antibody,anti-IL-33 neutralizing antibody and anti-TSLP neutralizing antibody respectively Then,the airway inflammation and airway remodeling were analyzed by HE staining and PAS staining of mouse bronchus;RT-PCR,Western Blot and immunohistochemistry were used to observe the expression of OPN in mouse lung tissue;ELISA was used to detect mouse serum,BALF The expression of OPN in asthmatic mice was comprehensively assessed,and the effect of anti-IL-25,IL-33,and TSLP neutralizing antibodies on OPN and airway remodeling in asthmatic mice were comprehensively assessed.Results:1.ELISA results showed that compared with the normal group,the expression of OPN in serum and BALF of the asthma model group was significantly increased(P<0.05);the anti-IL-25,IL-33,TSLP neutralizing antibody group Compared with the mice in the asthma model group,the expressions of OPN in serum and BALF were significantly decreased(P<0.05).2.Western blot and immunohistochemical results showed that the protein expression of OPN in the lung tissue of the asthma model group was significantly higher than that of the normal group(P<0.05);anti-IL-25,IL-33,Compared with the mice in the asthma model group,the protein expression of OPN in the lung tissue of the mice in the TSLP neutralizing antibody group was significantly decreased(P<0.05).3.RT-PCR results showed that compared with the normal group mice,the OPN gene expression value in the lung tissue of the asthma model group mice was significantly increased(P<0.05),and the anti-IL-25,IL-33,TSLP neutralized Compared with the mice in the asthma model group,the gene expression of OPN in the lung tissue of the mice in the antibody group was significantly decreased(P<0.05).4.The results of HE and PAS staining showed that compared with the normal group,the asthma model group had significantly more inflammatory cells in the bronchial wall(large nuclei,hyperchromatic,large cell volume,irregular),thickened tracheal wall,and goblet-shaped bronchial walls.Compared with the asthma model group,the asthmatic mice treated with anti-IL-25,IL-33,and TSLP neutralizing antibodies had significantly fewer inflammatory cells in the bronchial wall,thinner tracheal wall,and hyperplasia of goblet cells.Not obvious.5.The results of BALF count and classification showed that the count and percentage of eosinophils in the bronchoalveolar lavage fluid of the asthma model group were significantly higher than those of the normal group(P<0.05),anti-IL-25,IL-33 Compared with the mice in the asthma model group,the eosinophil counts and percentages of the mice in the,TSLP neutralizing antibody group were significantly decreased(P<0.05).Predominantly eosinophils.Conclusion:1.In bronchial asthma mice,the expressions of OPN,IL-25,IL-33 and TSLP in serum,BALF and lung tissue increased;2.Neutralizing antibodies against IL-25 and anti-IL-33 After the intervention of neutralizing antibody and anti-TSLP neutralizing antibody,the expression of OPN in serum,BALF and lung tissue decreased,and the airway remodeling of bronchial asthma was improved;3.In the airway remodeling of bronchial asthma mice,OPN and IL-25,IL-33,TSLP may have a certain relationship. |
