Network Pharmacology Analysis And Clinical Effect Observation Of Bushen Guchong Decoction On URSA Treatment Based On The Principle Of Preconditioning Its Loss

Objective:Network pharmacology was used to explore the potential targets and signal pathways of Bushen Guchong Decoction in improving unexplained recurrent spontaneous abortion(URSA).Based on the research results of network pharmacology,observing Bushen Guchong Decoction treatment under the guidance of "pre-peiqi damage" treatment Changes in TCM syndrome scores,basic endocrine hormones and cellular immune indexes in patients with kidney qi deficiency syndrome URSA,to discuss the mechanism and effectiveness of its intervention in the pre-pregnancy stage.Methods: 1.With the help of the Traditional Chinese Medicine System Pharmacology Database(TCMSP,http://tcmspw.com/tcmsp.php/)and the analysis platform and the bioinformatics analysis tool of the molecular mechanism of traditional Chinese medicine(BATMANTCM,http://bionet.ncpsb.org.cn/ batman-tcm /),screened the active compounds and targets in Bushen Guchong Recipe,and used Cytoscape 3.7.0to construct a traditional Chinese medicine-compound-target network map of Bushen Guchong Recipe;URSA disease was searched through Gene Cards,OMIM database and literature database Relevant targets;take the intersection of the drug targets of Bushen Guchong Recipe and URSA disease targets,and use String and Cytoscape to build a protein-protein interaction(PPI)network;finally,perform GO(gene ontology)on the intersection targets.and KEGG(Kyoto encyclopedia of genes and genomes)pathway enrichment analysis.2.60 cases of URSA patients with kidney-qi deficiency syndrome collected from the Affiliated Hospital of Shandong University of Traditional Chinese Medicine were randomly divided into an observation group and a control group,with 30 people in each group.Pre-pregnancy intervention was started on the 5th day of the menstrual cycle.The observation group took Bushen Guchong Recipe,and the control group took Cirong Yishen Granules.Both of them took 15 doses in a row for two menstrual cycles.The scores of TCM syndromes before and after treatment in the two groups were observed.,basal endocrine hormone estrogen(estradiol,E2),luteinizing hormone(luteinizing hormone,LH),follicle stimulating hormone(folliclestimulating hormone,FSH)levels and changes in cellular immune indexes were compared and analyzed.Results: Obtained 138 active compounds of Bushen Guchong Recipe,602 targets,537 known therapeutic targets related to URSA disease,and 84 intersecting targets.The biological processes obtained by GO enrichment analysis involve the response to hormones,the development of the immune system,etc.,the molecular functions involve the binding of tumor necrosis factor receptor superfamily,etc.,and the cellular composition involves membrane rafts,vesicle lumen,transcriptional regulatory complexes,receptors,etc.In terms of complexes,KEGG analysis showed that Bushen Guchong Recipe might regulate URSA status through AGE-RAGE signaling pathway,MAPK signaling pathway,TNF signaling pathway,PI3K-Akt and other signaling pathways.2.In the clinical study,after the intervention,the syndrome scores of kidney-qi deficiency in the observation group and the control group were both decreased,and the differences were statistically significant(P<0.05),the difference was statistically significant(P<0.05).After treatment,the FSH of the observation group and the control group were both lower than before,and the E2 of the observation group was higher than before,and the differences were statistically significant(P<0.05).After treatment,the CD3+T%,CD3+CD4+T%,CD4+/CD8+,NK cells(%)and B lymphocytes(%)in the observation group and the control group were decreased,and the differences were statistically significant(P<0.05),CD3+CD8+T% increased,and the difference was statistically significant(P<0.05).Compared with the control group,the CD3+CD4+T%,CD3+CD8+T%,CD4+/CD8+ of the observation group and the control group after treatment Compared with each other,the difference was statistically significant(P<0.05).Conclusion: With the help of network pharmacology analysis,it was shown that Bushen Guchong Recipe can improve URSA through anti-inflammatory,regulating immune balance,regulating blood state,etc.,with multiple targets and multiple pathways;clinical observation and analysis,URSA patients taking Bushen Guchong Recipe for conditioning during the prepregnancy stage can significantly improve the Patients with kidney qi deficiency symptoms,improve basic endocrine,and induce immune tolerance by reducing peripheral blood CD3+T%,increasing CD3+CD4+T%,and reducing CD4+/CD8+ ratio.
