Clinical Value Analyses Of IL20RB And UPS In Clear Cell Renal Cell Carcinoma

Background.Clear cell renal cell carcinoma(ccRCC)is a type of lethal malignant tumor of the urinary system.IL20RB is a cytokine receptor subunit coding gene and was found to play an important role in autoimmune diseases and some cancers,but its role in ccRCC is unknown.The ubiquitin proteasome system(UPS)controls protein degradation and plays a vital role in human tumors.We are aimed to make a comprehensive exploration of the clinical value of IL20RB and UPS in ccRCC.Methods.In the first part,we explored the prognostic value and therapeutic potential of IL20RB in ccRCC by various tools,including GSEA,R,Perl,UALCAN,GEPIA,cBioPortal,MethSurv,Xena,Esurv,TIMER,KM Plotter,TCGA,GEO,Oncomine,CCLE,Expression Atlas and STRING.We focused on the expression,prognosis value,clinical correlation,genetic alteration,methylation,function,and immune infiltration analyses to understand the clinical value of IL20RB in ccRCC.In the second part,the expression and clinical data of two ccRCC cohorts,TCGA-KIRC and GSE29609,were downloaded from TCGA and GEO database.We focused on the expression,prognosis value,clinical correlation analyses to understand the UPS’s clinical value in ccRCC.Further,we constructed two prognostic predicting models with UPS genes.Results.IL20RB is upregulated in ccRCC,and low methylation may be an underlying mechanism for its upregulation.Both high expression and low methylation of IL20RB predict worse survival,and both have a strong positive correlation with clinical features.Results suggest that IL20RB and neutrophils may interact with each other.Additionally,the immune microenvironment could influence the prognosis predicting ability of IL20RB.We finally identified 91 differentially expressed UPS genes and 391 prognosis-related UPS genes.Our two prognostic predicting models can indicate patients’ survival risk accurately both in TCGA-KIRC and GSE29609 cohorts.Conclusions.IL20RB plays a vital role in ccRCC,it could be a novel prognostic and potential therapeutic biomarker in ccRCC.UPS is dysregulated in ccRCC.UPS related genes have excellent prognostic value in ccRCC.Models constructed with UPS genes are effective and applicable.IL20RB and UPS have significant clinical value in ccRCC.
