| Research Objectives: The clinical literature on treatment of constipated irritable bowel syndrome with Traditional Chinese medicine in the past 40 years was reviewed,using data mining technology involved in the references related to syndrome differentiation type,traditional Chinese medicine prescription information for data analysis,at the same time,choose the highest correlation representative at the core of the drug combination,using technology to explore the treatment of traditional Chinese medicine network pharmacology action mechanism of constipation type of irritable bowel syndrome,It provides data support and documentation basis for clinical treatment and experimental direction of TCM treatment of constipation irritable bowel syndrome.Research methods: Using computers to search CNKI,VIP and Wanfang databases,search the clinical experimental literature on the treatment of constipation-predominant irritable bowel syndrome with traditional Chinese medicine in the past 40 years(retrieving date by 2020.11.01)experimental class literature.Excel2019 was used to statistically analyze the frequency of TCM use,the distribution of four qi and five flavors,meridian,efficacy and disease syndrome type;The ancient and modern medical case cloud platform was used to analyze the association rules of prescription Traditional Chinese medicine,and the complex network analysis of drug use of various syndromes was carried out.In the network pharmacology analysis,the active components and action targets of high frequency drugs against Bupleurum-paeony were collected through TCMSP database and related literatures.Gene Cards,OMIM and Drug Bank databases were used to obtain disease targets,and the intersection of disease and drug targets was analyzed by Venny2.1 software.The String database platform,David PPI network analysis was carried out on the overlap between targets and GO function and KEGG pathway enrichment analysis.Research results: were included in this study with 255 references,involving256 prescriptions,191 191 Chinese herbs,use frequency up to the top ten 191 Chinese herbs as: radix paeoniae alba,atractylodes,acid-insoluble ash,radix bupleuri,radix glycyrrhizae,betel nuts,fire maren,bitter orange,magnolia bark,wood.The top ten drugs in terms of efficiency and frequency were tonifying drugs,regulating qi drugs,purging drugs,relieving surface drugs,deworming drugs,dampening drugs,promoting blood circulation and removing stasis drugs,heat-clearing drugs,relieving cough,reducing phlegm and antiasthmatic drugs,and invigorating water infiltration drugs.In the prescription,warm drugs were the most used,followed by warm,mild cold,flat.In terms of medicinal taste,bitter,spicy and sweet drugs were used the most.The most drugs in the prescription were spleen meridian,followed by stomach meridian,lung meridian,etc.According to the association rule analysis,when the confidence level was100%,the top three association rules with support were Bupleurum-paeonia alba,glycyrrhiza-Paeonia alba and Atractylodes atractylodes alba.Complex network analysis obtained the core drugs for the treatment of IBS-C syndrome types,among which the core drugs for the treatment of heat and mimetic were:Ophiopogon,Rehmannia glutinosa,Magnolia officinalis,Hemp seed,Immaturus aurantii,Rhubarb,etc.The core drugs for the treatment of qi-mi include processed glycyrrhiza,Codonopsis pilosula,Rhubarb,xylobacter,glycyrrhiza,Atractylodes atractylodes,etc.The core drugs for the treatment of qi deficiency and secret are paeoniae alba,hemp,Bupleurum,prepared glycyrrhiza glycyrrhiza,Astragalus membranaceus,etc.The core drugs for the treatment of Yin deficiency and secret include radix scrophulariae,areca nut,paeonia alba,Radix Pseudostellariae,hemp seed and Radix ophiopogonis etc.The core drugs for the treatment of Yang deficiency and secret include Aconite,fructus aurantii,Bupleuri,Tuckahoe,licorice,angelica,etc.In the network pharmacology analysis,31 active components and 133 potential targets of Bupleur-paeonia lactiflora were obtained.The core targets of PPI analysis were AKT1,IL6,JUN,TP53,VEGFA and CASP3.The GO enrichment function involved 340 items,including the reaction to drugs,aging and other biological processes,enzyme binding,binding with protein and other molecular functions,extracellular space,membrane raft and other cellular components.133 KEGG Pathways were enriched,involving Pathways in cancer,Hepatitis B,TNF signaling Pathway and other related Pathways.Molecular docking results showed that the main active ingredients quercetin,kaempferol and isorhamnetin had strong binding effects with key target proteins AKT1,TP53 and IL6.Conclusion: The pathogenesis of IBS-C is complex and there are various syndromes.The drugs are mainly used to supplement deficiency,regulate qi and relieve diarrhea.The drug pairs of Bupleur-paeony are the most common in the prescription of IBS-C,which may act on key targets such as AKT1,TP53 and IL6 through active ingredients such as quercetin,kaempferol and isornetin.Through inflammatory,immune,endocrine and metabolic signaling pathways,ib S-C plays a therapeutic role.This study analyzed the medication rule of TCM in the treatment of constipated irritable bowel syndrome through data mining method,and revealed the material basis and molecular mechanism of Bupleurpaeony in the treatment of constipated irritable bowel syndrome through network pharmacology method,providing reference for clinical treatment and in-depth research of the disease. |
