| Objective:To explore the effect of the medicinal serum contained in tonic Yiqi soup on the proliferation of human laryngeal squamous cell carcinoma cells in in vitro experiments,the expression values of Lnc RNA UCA1 and mi RNA-185-5P in cells were determined,and the expression values of Lnc RNA UCA1 and mi RNA-185-5P were preliminarily explained that The tonic Yiqi soup inhibited the proliferation and invasion of LSCC cells through the Lnc RNA UCA1/mi RNA-185-5P signal axis.Methods:(1)Preparation of medicinal serum in Bu Zhong Yi Qi: SD rats with a body weight of 150 g ± 25 g were selected as experimental animals,and divided into blank group,normal saline group,low concentration group,medium concentration group,and high concentration group,and according to the body surface area,the Bu Zhong Yi Qi was taken from the abdominal aorta blood after 14 days of gradient gassing at a concentration of 1.05g/kg.(2)Passage and intervention of experimental cells: After the recovery of frozen human laryngeal squamous cell carcinoma cells FADU,according to the cell value-added situation,it is passed to the third generation of post-seed plates,and the medicinal serum of Bu Zhong Yi Qi is given in the logarithmic stage of cell growth,and it is divided into five groups according to the serum concentration,namely blank group,A natural saline group,5% Bu Zhong Yi Qi Buserum concentration group containing the drug,10% Bu Zhong Yi Qi serum concentration group containing the drug,and 20% Bu Zhong Yi Qi serum concentration group containing the drug.(3)CCK8 detects cell activity and value-added(4)q PCR detects the expression of two genes of interest in cells.Finally,analyze the data.Results:The determination of Lnc RNA UCA1 shows that the expression value of each tonic Yiqi soup drug-containing serum group decreased compared with the control group,and the experimental group was different from the control group,and the p<0.05,in addition,the 20%tonic Yiqi soup drug-containing serum group was compared with the 5%tonic Yiqi soup drug-containing serum group,p <0.05。 However,there was no difference in the expression of Lnc RNA UCA1 between the 5%tonic Yiqi Soup drug-containing serum group and the 10% tonic Yiqi Soup drug-containing serum group,and there was no difference between the 20% tonic Yiqi Soup drug-containing serum group and the 10% tonic Yiqi Soup drug-containing serum group.The determination of mi RNA-185-5P shows that after the intervention of the drug-containing serum of Tonic Yiqi Tang,the expression of mi RNA-185-5P also gradually increased when the serum concentration increased,and there was a characteristic of concentration dependence.After the one-way ANOVA data,there were differences between the experimental group and the control group of Tonic Yiqi Soup,and there were also differences between the three intervention groups with different concentrations,p<0.05.Conclusion:The medicinal serum containing Yiqi soup can inhibit the proliferation of laryngeal squamous cell carcinoma cells through the Lnc RNA UCA1/mi RNA-185-5P axis,and the possible mechanism is to downregulate the expression of Lnc RNA UCA1 and improve the expression of mi RNA-185-5P. |
