Objective By detecting the expression level of fibroblast activation protein(FAP)in the kidney tissue of renal interstitial fibrosis(RIF)rats,the role of FAP in the occurrence and development of RIF was explored.Method Forty male SD rats(120-150 g)were randomly divided into four groups: sham operation group,model group,inhibitor group and negative control group,with 10 rats in each group.Rats in the sham operation group did not ligate the ureter after laparotomy;rats in the model group found the left ureter to be ligated and severed after laparotomy;Rats in the inhibitor group and the negative control group also found the left ureter ligated and severed after opening the abdomen,and were injected with FAP inhibitor(UAMC-1110)and normal saline into the tail vein every other day one week later,a total of three times.All the rats were sacrificed 14 days later,and the left kidney section of the rats was taken for HE staining and Masson staining to detect the pathological changes of the kidney tissue,and calculate the rat kidney RIF index.Immunohistochemical method was used to detect the protein expression of FAP,Transforming growth factor-β1(TGF-β1)and α-Smooth muscle actin(α-SMA)in rat kidney tissue,Real-time fluorescence quantitative polymerase chain reaction(RT-q PCR)was used to detect the m RNA expression of FAP,TGF-β1and α-SMA in rat kidney tissues,Western blotting(WB)was used to detect the protein expression of FAP,TGF-β1 and α-SMA in rat kidney tissue.Results 1.Compared with the sham operation group,the RIF index of rats in the model group was significantly increased(P<0.05);compared with the model group,the RIF index of rats in the negative control group had no significant difference(P>0.05),while the inhibitor group The RIF index of rats was significantly reduced(P<0.05).2.Compared with the sham operation group,the m RNA and protein expressions of FAP,TGF-β1 and α-SMA in the kidney tissue of the model group were significantly increased(all P<0.05);compared with the model group,the negative control group was larger There was no significant difference in the m RNA and protein expression of FAP,TGF-β1 and α-SMA in rat kidney tissues(all P>0.05);compared with the model group,the inhibitory group had a significant difference in renal tissue FAP,TGF-β1 and α-SMA.Both m RNA and protein expression were significantly reduced(both P < 0.05).Conclusion inhibition of FAP α expression may reduce RIF in rats with unilateral ureteral obstruction.3.Correlation analysis: FAP in rat kidney tissue was significantly positively correlated with TGF-β1 and α-SMA protein expression and RIF index(r=0.964,0.956,0.968,all P<0.05).Conclusion In UUO-induced RIF rats,the high expression of FAP in kidney tissue may be involved in the occurrence and development of RIF;inhibiting the expression of FAP can reduce the degree of RIF in rats.
