| Objective:The purpose of this project is to systematically study the quality tandards of the Yao drug Cyclea Hypoglauca(Schauer)Diels,and to formulate a strongquality evaluation method for it.Clinical efficacy of Cyclea Hypoglauca(Schauer)Diels for the treatment of pharyngitis,tonsillitis,enteritis,and inflammation is likely to be caused by bacteria,the anti-inflammatory effect,antibacterial effect,acute toxicity and other pharmacological contents of the clinical application were preliminarily explored,so as to provide a basis for the pharmacological activity study of the clinical application.Methods:1.The base source,characters,microscopic characteristics,chemical identification and TLC identification of Cyclea Hypoglauca(Schauer)Diels were carried out in-depth and meticulous dentification.2.According to a series of requirements in the Chinese pharmacopoeia,the water content,total ash content,acid insoluble ash and four kinds of leaching objects of different batches of Cyclea Hypoglauca(Schauer)Diels were measured.3.The optimum extraction method of total alkaloids from Cyclea Hypoglauca(Schauer)Diels investigated by using the Orthogonal experimental design,and the content of total alkaloids indifferent batches of Cyclea Hypoglauca(Schauer)Diels samples was determined by the UV.4.Oxford cup method and double dilution method were used to study the in vitro antibacterial effect of Cyclea Hypoglauca(Schauer)Diels,and the minimum inhibitory concentration(MIC)was determined.5.An experimental model of auricle swelling was used to study the inhibition of acute inflammation in mice.6.According to the relevant standards and requirements of acute toxicity test,the toxicity and the maximum tolerable dose of Cyclea Hypoglauca(Schauer)Diels to mice were determined.Results:1.The microscopic structure of cross section,chemical identification and TLC can be used for qualitative identification.2.To determine the water content of Cyclea Hypoglauca(Schauer)Diels,the water content does not exceed 10.0%.To determinethe total ash content of Cyclea Hypoglauca(Schauer)Diels,the total ash content should not exceed10.0%.To determine the acidinsoluble ash content of Cyclea Hypoglauca(Schauer)Diels,the content of acid insoluble ash should not exceed 1.50%.The content of water leaching isn’t less than 4.00%,and the content of cold alcohol leaching isn’t less than 2.00%.3.The optimum extraction condition of total alkaloids was 80℃,the ethanol concentration was 50%,the material ratio was 1:60,and the extraction time was90 min.The total alkaloids content is between 3.52%~5.56%.4.Cyclea Hypoglauca(Schauer)Diels can inhibit Staphylococcus aureus,Candida albicans,Bacillus subtilis,Pseudomonas aeruginosa,Proteus and Enterococcus faecalis to different degrees.5.Cyclea Hypoglauca(Schauer)Diels has anti-inflammatory effect on acute inflammation induced by xylene.6.The lethal 50% dose could not be measured,and the maximum tolerance of mice to Cyclea Hypoglauca(Schauer)Diels was 40g/kg,greater than 5g/kg,which proved that drug was non-toxic.Conclusion:1.The characteristics of cross section,microstructure,chemical identification and TLC can be used as the main basis for the identification.2.The water content,total ash content,acid insoluble ash content and extract content can reflect the quality of the Cyclea Hypoglauca(Schauer)Diels to a certain extent.3.Using the Palmatine as reference,it is feasible to use UV spectrophotometry to determine the total alkaloids of Cyclea Hypoglauca(Schauer)Diels,and the total alkaloids can be used as the evaluation index of the quality of Cyclea Hypoglauca(Schauer)Diels.4.Cyclea Hypoglauca(Schauer)Diels has antibacterial and anti-inflammatory effects and is non-toxic. |
