Objective:To compare and analyze the differences in microRNA expression between patients with coronary atherosclerotic heart disease and healthy people,and miRNA expression between patients with stable CAD plaques and patients with unstable plaques,and to explore the possible effects of differentially expressed miRNAs on CAD and plaque stability.Method:Ten healthy people,ten patients with CAD were enrolled by this study.coronary angiography after admission using optical coherence tomography to assess coronary artery disease,plaque stabilization group and divided into groups according to the presence or absence of plaque instability vulnerable plaque,from plasma Separate exosomes,use electron microscopy,particle size analysis,protein and fluorescent labeling,perform miRNA extraction and concentration measurement,and evaluate the relationship between the differential expression of circulating miRNA and CAD and the stability of plaques.Finally,the miRNA database is used to predict the target gene,and its related pathways are further analyzed through functional enrichment analysis,annotation analysis and protein interaction network.Results:(1)10 differentially expressed miRNAs were detected in the plasma of healthy subjects and CAD humans,miR-532-5p,miR-629-5p,miR-409-3p,miR-425-5p,181a-2-3p,let-7g-5p,miR-148b-3,miR-186-5p,miR-484,miR-10395-3p were significantly down-regulated in patients with coronary heart disease.(2)In the AS plaque stable group and the unstable group,five differentially expressed miRNAs were found,of which miR-4446-3p was up-regulated in the plaque unstable group,miR-375-3p,miR-28-3p,MiR-193a-5p and miR-92a-3p were down-regulated in the unstable plaque group.(3)The functional enrichment of differentially expressed miRNAs in CAD patients is mainly autophagy mechanism,protein breakdown,histone modification,and related pathways mainly involve neurodegeneration,cell aging and autophagy.The functional enrichment of differentially expressed miRNAs in patients with stable plaques and patients with unstable plaques is mainly proteolysis and histone modification,and related pathways mainly involve cell cycle,cell aging and so on.(4)The differentially expressed miRNAs of CAD patients and healthy people and the differentially expressed miRNAs of the CAD plaque stable group and the plaque unstable group did not find the common differentially expressed miRNA.Conclusion:Significantly differentially expressed exosomal miRNAs were found between CAD patients and normal healthy people,between stable plaque groups and unstable plaque groups.Most of these miRNAs are related to inflammation and protein receptor-related functions and pathways.It is also closely related to CAD,and further study of these differentially expressed miRNAs is of great significance for further understanding of CAD.
