| Dang gui Bu xue decoction(DGBXD)has a certain effect in anti-tumor therapy.Angelica sinensis polysaccharides(ASP)and Astragaloside IV(AST)are one of its active ingredients.The efficacy of traditional preparations is usually affected by poor patient compliance,hydrophobicity and non-selective distribution and so on.In order to overcome the above shortcomings,ASP was is used as carrier to load AST,and the ASP-AST-magnetic nanoparticles(MNP)(ASP-AST-MNP)is prepared for anti-tumor therapy by this topic.In order to explore the synergistic effect of the multi-component traditional Chinese medicine with drug-loaded nanoparticle preparations,and through in vitro cell experiments,preliminary evaluation of its in vitro effect on breast cancer.The phenol-sulfuric acid method was used to establish a method for the determination of sugar content,and the o-phenanthroline method was used to establish a method for the determination of iron content.The ASP-magnetic nanoparticles(ASP-MNP)were prepared by co-precipitation method,and the pharmaceutical properties of ASP-MNP were investigated.hese contents of the above research can be used as the basis of previous research.ASP-AST-MNP was prepared by double emulsion solvent evaporation method.Through single factor inspection to determine the best preparation conditions.The drug-loaded nanoparticles were characterized by particle size,polydispersity index(PDI),zeta potential,magnetic response,entrapment efficiency and drug loading,and their sugar content,iron content,AST content and stability were determined.Taking human breast cancer MCF-7 cells as model cells,MTT method was used to evaluate the anti-tumor effect of human breast cancer MCF-7 cells in vitro.The magnetic targeting of ASP-AST-MNP was investigated by Prussian blue stainingThe ASP-MNP was prepared by coprecipitation method,to determine the feasibility of ASP as nano drug carrier.The ASP-AST-MNP was prepared by double emulsion solvent evaporation method,and the methodological investigation met the requirements.The content of AST in the prepared magnetic nanoparticles was(0.98±0.03)μg·μL-1,drug loading was(10.86±0.36)%,entrapment efficiency was(78.40±2.40)%.The stability of the prepared nanoparticles was good.The results of cytotoxicity test showed that the inhibitory rate of ASP-AST-MNP on human breast cancer MCF-7 cells in vitro was significantly higher than that of ASP group and AST group.In addition,the inhibitory effect of ASP-AST-MNP on cells was dose-dependent,and the cell inhibition rate increased significantly with the increase of concentration.The ASP-AST-MNP prepared by Prussian blue staining showed that the nucleus was stained blue,demonstrating that ASP-AST-MNP has been ingested by cells.In the magnetic targeting experiment,the cells that absorbed the magnetic nanoparticles by external magnetic action had obvious magnetic targeting.The findings of this study show that it can be that it is feasible to prepare magnetic nanoparticles by using ASP as carrier to load AST.The ASP-AST-MNP is successfully prepared.The in vitro cytotoxicity test confirmed that ASP-AST-MNP has obvious cell inhibition rate.Prussian blue staining confirmed that the magnetic nanoparticles had been ingested by the cells,and had good magnetic targeting under the action of the magnetic field.This subject used the combination of multi-components of traditional Chinese medicine and modern nanotechnology to prepare magnetic nanoparticles with synergistic effect,which fully reflects the application value of traditional Chinese medicine preparation in targeted drug delivery system. |
