Objective:The purpose of this paper was to observe the effects of low-requency low-intensity ultrasound(LFLIU)combined with curcumin on proliferation,migration and expression of chemokine receptor CXCR4 in human glioma U87 cell line.It will analyze whether the effect is more effective by using LFLIU and curcumin in combination down-regulate the expression of CXCR4.Methods:CCK-8 was used to measure the proliferation of U87 cells after treated with the curcumin at concentration of 0,5,10,and 20μmol/L respectively,with the reaction time of 24 h to U87 cells.CCK-8 was used to measure the proliferation of U87 cells after treated with the LFLIU at intensity of 0,50.4,83.4,142,and 290 m W/cm2,irradiation time of 60 s.We get the ultrasound and curcumin combined treatment parameters,which is intensity of 142.0m W/cm2 and concentration of 10μmol/L.Dectecting the effects of curcumin and ultrasound alone or in combination on the cell migration ability by transwell migration assay.Dectecting the effects of curcumin and ultrasound alone or in combination on the expressions of CXCR4 proteins by western blotting analysis.Results:1.Curcumin alone significantly inhibited proliferation of the U87 cells in a dose-dependent manner.Curcumin concentration at 5μmol/L had no significant proliferation inhibition compared with the control group(P>0.05),and the concentration of curcumin is 10,20μmol/L could inhibit the U87 proliferation significantly(P<0.05).2.Low-frequency low-intensity ultrasound alone significantly inhibited proliferation of the U87 cells in an intensity-dependentmanner.Ultrasound with intensity≤50.4m W/cm2 did not inhibit the U87 cells proliferation compared with the control group(P>0.05).Ultrasound with intensity≥83.4m W/cm2 could inhibit cell proliferation significantly(P<0.05).3.Proliferation inhibition effect of combined group(C10U142.0)is more significant than the control group(P<0.05),and proliferation inhibition effect of combined group more significant than that of curcumin and low frequency low intensity ultrasound alone(P<0.05).4.The inhibition cell migration ability of combined group(C10U142.0)is more significant than the control group the single group(P<0.05).5.LFLIU(142.0m W/cm2)combined with curcumin(10μmol/L)could down-regulate the expression of CXCR4 protein,and the expression in combined group was significantly higher than that of the single treatment group(P<0.05).Conclusion: LFLIU and curcumin alone could inhibit the proliferation and migration,and down-regulate the expression of CXCR4 in human glioma cells,and the combination of LFLIU and curcumin had synergistic effect. |
