Objective Research of aconite-rhubarb drug pair on the therapeutic effect of kidney Yang deficiency syndrome,and the influence on the hypothalamus-pituitary-adrenal axis,to explore its possible treatment mechanism and provide experimental basis for clinical using.Methods1.Using the ultrasonic extraction method to extract the effective components in the aconite – rhubarb and using the HPLC to determinate the content of mesaconitine,hypaconitine and aconitine the extract,explore the changes in content of three diester-type alkaloids in aconite after compatibility with rhubarb.2.The 60 male SD rats were randomly divided into 6 groups,each group of 10,including the control group,the model group,the aconite-rhubarb(1:1)group,the aconite-rhubarb(1:3)group,the aconite group,and the rhubarb group respectively.In addition to the control group,the remaining was administrated hydrocortisone(25mg/kg,HYD)for 14 days by i.m,and in control group hydrocortisone was replaced with saline(2ml).Then the kidney Yang deficiency rat model was made.From the 15 th day in experiments,1:1,1:3,aconitine,rhubarb group respectively were given of aconite-rhubarb(1:1,3 mg/kg),aconite-rhubarb(1:3,6mg/kg),aconite(1.5 mg/kg),rhubarb(1.5 mg/kg)for 21 days by gastric gavage,control group and model group were replaced with saline(4 ml).During the experiment,the rats were weighed once every 2 days and measured body temperature once weekly.On the 36-day in the experiment,rats were hocussed with 1% pentobarbital sodium,and then collected blood(2ml)from abdominal aorta to detect c Amp c Gmp and Cort in serum,the hypothalamus and pituitary of rats were used to detect the content of CRH and ACTH,using immunohistochemical method to detect the content of GR and fluorescence quantitative PCR method to detect GRm RNA expression in adrenal gland.Results1.After the combination with rhubarb,content of three diester-type alkaloids were significantly lower than those in the separate group.2.Compared with the control group,model group and rhubarb group weight and body temperature decreased significantly(p < 0.05),c AMP and the c AMP/c GMP ratio in serum declined significantly(p < 0.05)and a marked increase in the c GMP,the content of Cort also significantly reduced at the same time(p < 0.05),the viscera of CRH,ACTH,GR and GRm RNA levels were significantly lower(p < 0.05),after giving 1:1 and aconite,can effectively alleviate the multi-index abnormalities,and the effects on 1:1 on body weight,body temperature,the c AMP/c GMP ratio,CRH,GR,GRm RNA were better than 1:3(p <0.05),the influence of 1:1 on ACTH and GRm RNA also superior to separate groups of aconite(p < 0.05).Conclusion After the compatibility with rhubarb,aconite can effectively reduce the extract content of three kinds of diester-type alkaloids and this drug pair can effectively enhance treatment effect of kidney Yang deficiency syndrome,its main mechanism related to effects on the function of HPA axis.
