| Objective:The electrochemical conditions for the detection of heavy metals and harmful elements in traditional Chinese medicine were optimized.The method of rapid detection Cd,Pb and Cu of elements in Astragalus membranaceus were studied to provide basis for real-time monitoring of the quality of Chinese herbal medicines of a new methods and pollution status.In addition,ICP-MS for the detection of heavy metals and harmful elements have been established in the complex matrix of traditional Chinese medicines.It can be used to determine the concentration of Astragalus membranaceus that may cause heavy metals and harmful elements pollution in the entire industrial chain.And we conducted a health risk assessment study on pollution levels in conjunction with relevant international and domestic standards.Method:(1)Three kinds of electrochemical techniques(SWASV,DPV,LSV method)were used to compare Cd,Pb,and Cu of the oxidation-reduction peak potentials,the peak current intensity,peak shape symmetry and baseline smoothness to optimize the best detection technology and corresponding parameters.In addition,the optimal ion deposition potential of heavy metals were optimized by using SEM to observe the deposition morphology of harmful elements,and combined with electrochemical detection accuracy and reproducibility test.(2)SWASV method was established to detect the linear regression equations of Cd2+,Pb2+,and Cu2+in the aqueous phase and the matrix of Astragalus membranaceus and conducting methodological investigation.The contents of Cd2+,Pb2+ and Cu2+ were detected to the 10 batches of Astragalus membranaceus and the results were verified by ICP-MS.(3)The ICP-MS method was established to detect the standard processes and methods for Cr,Cu,As,Cd,Hg,and Pb of the heavy metals and harmful elements in the matrix of Astragalus membranaceus.These factors were evaluated to affect the quality and safety of Astragalus membranaceus that the planting environment,medicinal sites,growing years,planting areas,and processing methods and so on.Result:(1)The SWASV method was used to optimize the test conditions for depositing potentials of-1.1V(Cd2+),-1.0V(Pb2+)and-0.6V(Cu2+),and a deposition time of 120s.(2)A method was established to detect Cd,Pb,and Cu of the harmful elements in Astragalus membranaceus.This method is efficient,fast,and reproducible.This method was used to rapidly screen the contents of Cd2+,Pb2+ and Cu2+ in 10 batches of commercially available Astragalus membranaceus samples,and the results were consistent with those of ICP-MS.(3)Based on the reference material of Astragalus membranaceus,the ICP-MS method was used to detect six kinds of heavy metals and harmful elements in Astragalus membranaceus with low detection limit and high precision.The content of Astragalus membranaceus in the whole industry chain were determined.The planting environment of Astragalus membranaceus had a significant relationship with the enrichment of heavy metals and harmful elements.The root safety of Astragalus membranaceus was higher than that of other parts.As the planting time increased,the concentrations of Cr,As,Cd,Pb were gradually increased in Astragalus membranaceus.The concentrations of heavy metals and harmful elements in Astragalus membranaceus in Inner Mongolia Autonomous Region were the highest,followed by Gansu and Shanxi.The concentrations of all elements in processed Astragalus membranaceus have declined.According to the analysis of health risk assessment data,Cr,Cu,As and Pb of heavy metals and harmful elements in Astragalus membranaceus have certain potential risks for the human body.Conclusion:The real-time monitoring of the development of electrochemical methods for the determination of Cd2+,Pb2+and Cu2+in Astragalus membranaceus will provide a basis for the quality control and safety of Chinese medicines.ICP-MS method was established to detect the contents of Cr,Cu,As,Cd,Hg,and Pb of six kinds of heavy metals and harmful elements in Astragalus membranaceus.The influencing factors for the quality of Astragalus membranaceus were determined to conducte the health risk assessment.This will provide a certain reference and guiding significance for the relationship between cultivation,processing,production process on heavy metals and harmful elements of Astragalus membranaceus. |
