| Objective:Pesticide carbendazim was the target of this study.Based on the principle of immunoassay,a colloidal gold immunochromatographic strip for rapid detection of carbendazim in traditional Chinese medicine was established.The application of the colloidal gold immunochromatography paper to different medicinal parts of medicinal materials was preliminarily investigated.Rapid screening of carbendazim in 53 batches of tangerine samples by colloidal gold immunochromatography paper strips.A super high performance liquid phase tandem mass spectrometry was established for the quantitative determination of carbendazim residues in 53 batches of tangerine samples,and the test results of test paper were confirmed.Method:The colloidal gold solution with suitable particle size was prepared by by sodium citrate method.The parameters and quality of colloidal gold solution were investigated by ultraviolet spectrophotometry and particle size scanning.Based on the specific reaction of antigen-antibody,a colloidal gold immunochromatographic test paper which can be used for rapid detection of carbendazim was developed.The method applicability of 54 Chinese medicinal materials in different parts of was investigated.The applicability of the test paper was preliminarily determined.A suitable extraction method was established,and the real samples were detected by the test strip.The accuracy of the test strip was confirmed by ultra high performance liquid chromatography tandem mass spectrometryResult:The colloidal gold particles with the average particle size of about 20nm were obtained by the reduction method.The solution showed bright wine red and the quality was stable.Through experimental comparisons,the most suitable pH and protein amount for the coupling of colloidal gold and carbendazim antibody were determined,and the stable gold labeled antibody was obtained.The assembly materials and preparation conditions of colloidal gold immunochromatography paper were optimized,and the optimum assembly materials and the best preparation methods were determined.When the residue of carbendazim is between 10 and 30 ng/mL,the T-line color is weaker enough than the C-line color with judgement by naked eyes.While residueis above 30 ng/mL,the T-line was absolutely disappeared.The application of the colloid gold gold immunochromatography paper was investigated.It was preliminarily proved that the test paper was suitable for 46 kind of.traditional Chinese medicine.The carbendazim residue in 53 batches of dried tangerine peel were detected by the colloidal gold immunochromatography and ultra performance liquid chromatography tandem mass spectrometry detection,The results show that the results of the two methods were highly consistent.The accuracy of colloidal gold immunochromatography paper method was confirmed.Conclusion:A colloidal gold immunochromatographic strip for rapid screening of carbendazim in Chinese traditional medicine was developed successfully.The detection area were less than 0.1 mg/kg,0.1~0.3 mg/kg and higher than 0.3 mg/kg.The visual limit of detection(LOD)was 0.1 mg/kg.The method is sensitive,specific,accurate,reliable,easy to operate,and less than 5 min within the detection time.It is suitable for large-scale on-site rapid screening,and can be used as a field monitoring and screening technology for carbendazim residues in traditional Chinese medicine. |
