Objective To investigate the inhibiting effect and the possible mechanism of puerarin on βamyloid peptide1-42-induced tau protein hyperphosphorylation in SH-SY5Y cells and rats.Methods Morris Water Maze is used to detect learning and memory ability of rats,ELISA and Western blotting is used to detect Aβ40、Aβ42,the protein expression of tau phosphorylation,and the protein level of GSK-3β,GSK-3β-Ser9、β-catenin、CyclinDl,the key factors of Wnt/(3-Catenin Signaling Pathway in SH-SY5Y cells and rats.Results Puerarin can signally enhance the ability of learning and memory in rats,decrease Aβ40、Aβ42,inhibite the level of tau phosphorylation at Ser396、Ser199、Thr231 induced by β-amyloid peptide1-42 in SH-SY5Y cells and rats,and can restrain the viability of GSK-3β by activating the expression of GSK-3β-Ser9,and increase the protein expression of P-catenin、CyclinD1,the key factors of Wnt/β-Catenin Signaling Pathway.Conclusion Puerarincan attenuate β-amyloid peptide1-42-induced tau protein hyperphosphoryl-ation in SH-SY5Y cells and rats,by inhibiting the exprssion of GSK-3β,and activating the Wnt/β-Catenin Signaling Pathway. |
