Objective To improve the multi-components detection method of steroids which could be used in clinical study.The metabolic characteristics of the steroids with varied ages and menstrual cycles were explored with the method.Then it was used for the study of the discoveries of the steroids which were significantly related to the development of Sj(?)gren’s Syndrome(SS).The exploration of the role of the steroids in the pathogenesis of SS may supply a reference for clinical study.Methods 1)The process of the pretreatment and analysis of the samples were standardized and fulfilled.And a method of the multi-components detection of steroids was established-2)The serum of the healthy normal women aged from 40 to 60 were collected.To explore the optimal detection interval of the samples,the quantitative detection of the steroids were analyzed with the platform.The relevance of ages/postmenopause and the fluctuations of the steroids was analyzed.3)50 cases of Sjogren’s syndrome(SS)patients(SS group)and 50 cases of healthy samples(healthy group)were included in this study.The steroids of the samples in this study were measured with quantitative detection via the platform.Results 1)The 13 steroids calibrators showed a linear response from 0.05 to 20 ng/mL(R2>0.995 6),with limit of detection was no more than 1 ng/mL,respectively;cortisol(CORT)and dehydroepiandrosterone sulfate(DHEAS)calibrator showed a linear response from 50-2000 ng/mL(R2>0.997 9),with limitation of detection was no more than 0.5 ng/mL.2)The results of the detection of the steroids during the whole menstrual cycle shows that the steroids with the similar properties fluctuated with the similar trends,such as P4 and 17-OHP in estrogens,Cort and S in cortical steroids,and T and Adione in androgens had the same performance.The mean percentage of the 15 steroids fluctuated with different properties,in which androgens reached the peak in late follicular phase,progesterone(estrogens)reached the peak in luteal phase,while the changes of cortical steroids were not so evident.3)The results of PLS-DA showed that SS group and healthy group can be well classed by 6 androgens.The levels of testerone(T),Dihydrotestosterone(DHT),dehydroepiandrosterone(DHEA),androsterone,and DHEAS in SS group were significant lower than those in healthy group(t=8.536,2.438,3.172,4.158,4.489,all P<0.05).The samples of premenopausal and postmenopausal women can be distinguished between two groups by PLS-DA.The levels of T and androsterone in SS group were significantly different from the healthy group in the premenopausal women(P<0.05),while besides T and androsterone,the levels of DHT/DHEA/DHEAS in SS group were significantly different from healthy group(P<0.05).Conclusions The study improved the multi-components detection method of steroids which could be used in clinical study,and was used to study the dynamic changes of the steroids in the whole menstrual cycle of women.Finally it was used to explore the difference between the women after menopause in SS groups and healthy groups.T and androsterione were the common steroids whose levels were significantly different in these two groups among all the women in this study.The decrease of androgens,may the leading cause of morbidity in postmenopausal women than in premenopausal women...
