Painengda Capsule’s Influence On Transcriptional Regulation Factor Of Th1/Th2 Cells In Rats With Hashimoto Thyroiditis

Objective:To research the influence of Painengda capsule on thyroglobulin antibody(TgAb),thyroid peroxidase antibody(TPOAb)levels and the pathological ultrastructure changes of the thyroid tissue observed by the transmission electron microscope in the rat model with experimental autoimmune thyroiditis(EAT).Preliminary discussion on this drug’s influence on transcriptional regulation factor of Th1/Th2 cells and T-bet,GATA-3 in the EAT.Methods:Healthy Wistar rats 40,all female,were randomly divided into five groups,8 for each group,normal group,the model group,low dose group of Painengda capsule,middle dose group of Painengda capsule and high dose group of Painengda capsule.The model group and Painengda capsule low,medium and high dose groups used Porcine Thyroglobulin(PTg)and Complete Freund’s Adjuvant(CFA)or Incomplete Freud’s Adjuvant(IFA)subcutaneous injection for six weeks to get the EAT model.After eight weeks treatment with concoction made by Painengda capsule,while the model group were treated only with saline,ELISA enzyme league immune detected rat serum autoantibodies TgAb and TPOAb levels.Then,all rats were executed and thyroid tissue were stripped to observe the pathological ultrastructure changes of thyroid tissue by the transmission electron microscope,RT-PCR were used to detect the mRNA of T-bet and GATA-3 in thyroid tissue.Results:1.The model group’s rats serum TgAb,TPOAb level were higher significantly than that of the normal group,and the two groups had significant difference(p<0.01),each Painengda capsule treatment group’s rats serum TgAb,TPOAb level were lower significantly than that of the model group,Low and Middle Painengda capsule treatment group had significant difference(p<0.05).2.Compared with the normal group,more severe pathological damage of thyroid follicular cell was observed by the transmission electron microscope.Compared with the model control group,each Painengda capsule treatment group’s thyroid follicular cell pathological damage under the transmission electron microscope was better,Low and Middle Painengda capsule treatment group had obviously improve among them.3.Compared with the normal group,the T-bet protein and mRNA expression level in model control group rats is significant increase(p<0.01).Compared with the model control group,each Painengda capsule treatment group’s rats had lower T-bet mRNA expression level,Low and Middle Painengda capsule treatment group had significant difference(p<0.05).The mRNA expression level of GATA-3 were higher in each Painengda capsule treatment group’s rats,Low and Middle Painengda capsule treatment group had significant difference(p<0.05)。Conclusion:1.Painengda capsule has certain treatment function to EAT,it could significantly reduce the TgAb and TPOAb levels in rats serum.2.Painengda capsule could significantly improve the injury of thyroid follicular cell in EAT rats.3.Painengda capsule could increase the mRNA expression level of GATA-3,decrease the mRNA expression level of T-bet.T-bet mRNA expression level rise is likely to be one of the important reasons which cause Hashimoto’s thyroiditis.
