| Cancer has become the most dangerous disease that threats people’s health in our country.People pay a lot of attention to it.According to statistics,compared to the first half of the 20 th century,global cancer rates is nearly 11 times in the second half of the 20 th century.And in our country,up to 2.5 million people died of malignant tumor.Although humans have invested a lot of money in prevention of tumor,the progress is still limited.So far,there is still no effective radical treatment for malignant tumor.Reducing cancer mortality needs to rely on early diagnosis and early treatment.Because early treatment will create more chance for tumors to survive,it is the key for tumor cure to detect tumor and give treatment as early as possible.Tumor markers is the important examination index for tumor patients.For some kind of tumor detection,targeted tumor markers with strong specificity are always chosen.For ordinary health check-up subjects,it is better to choose common tumor markers.Thus malignant tumor detection rate will be improved.Studies have shown that serum p53 antibody has close correlation with common malignant tumor,p53 gene mutation may occur in the early period of cancerous tumor patients,and encode the mutations of p53 protein,and induce the body to give immune reaction,and produce p53 antibodies.the antibodies usually have existed not long after malignant tumor appear.It’s convenient to make the antibody detection,and because it has high specificity,early serum p53 antibody detection can be used as a tool to find the tumor.Therefore,p53 antibody can be used as a broad spectrum of tumor markers for regular check-up subjects during tumor screening.This research uses restructured p53 protein and has phage SS and SP which show the advantages of p53 protein as envelope antigen.The research also uses experiment enzyme-linked immunosorbent(ELISA)in screening serum p53 antibody in 2954 cases of check-up subjects.According to the contents and changes of serum p53 antibody to predict the possibility of the tumor,and thus it will be achievable to realize the goal of early screening of tumor.At the same time,this study carried out AFP,CEA detection in 2954 cases of check-up subjects.And analyzed the correlation between p53,AFP,and CEA.This study carried out in 2011 tracked p53 antibody positive patients,exploring the correlation between p53 antibody abnormalities and the formation of tumors.The results of the study are as follows:1)A total of 103 p53 antibody positive patients.Among them,restructured p53 protein were detected in 79 cases(detection rate is 2.67%),comprising 76.5% of all the p53 antibody positive patients.double show phage package is detected in 72 cases(detection rate is 2.44%),comprising 69.9% of all the positive patients.48 patients appear both in restructured p53 protein detection cases and double show phage detection cases;2)Among the 103 cases of p53 antibody positive patients,55 patients have symptoms related to tumor except the other 26 cases.It suggests that p53 antibodies are a closely tumor marker 3)In the 103 p53 antibody positive patients,11 cases are CEA positive patients,and 2 patients are AFP positive.Software analysis showed that p53 antibody was significantly associated with the exception of the CEA(P = 0.015),Providing a reference basis for clinical tumor markers of united detection.4)After tracking the p53 antibody positive patients,we find that the rise of p53 antibody is closely related to the occurrence of tumor.Based on the technology of phage display,p53 antibody screening may provide a new approach to the discovery of early tumor detecting.It is expected to have reference value for screening of clinical tumor. |
