The Tibetan pig is a endemic pig species,mainly living in the Tibetan Plateau and its adjacent alpine and subalpine areas.According to the morphological and histological changes of pig ovaries,the estrus cycle of pig is usually divided into follicular phase and luteal phase.LncRNA plays an important role in many biological processes such as cell cycle regulation,cell differentiation regulation,epigenetic regulation and dose compensation effect in vivo.Therefore,by screening and analyzing the characteristics and differences of lncrnas in ovarian follicular stage and luteal stage of Tibetan pigs,lncrnas in ovarian tissue that affect the reproductive performance of Tibetan pigs can be identified,which can provide theoretical basis for promoting the healthy,stable,efficient and sustainable development of Tibetan pig breeding industry in Tibetan areas.In this experiment,the ovarian tissues of 6 5-month-old healthy Tibetan sows were selected and divided into two groups: follicular stage and luteal stage.Paraffin sections were taken for HE staining observation,and the structures of follicle cavity,cumulus and corpus luteum of follicle stage were observed,thus confirming the accuracy of grouping.A total of1055 differentially expressed lncrnas were found in the ovaries of Tibetan pigs at follicular stage and luteal stage by high-throughput transcriptome sequencing technology.Compared with the follicular stage group,532 lncrnas were up-regulated and 523 down-regulated in the luteal stage group.At the same time,the target genes of co-expressing LncRNA in ovarian tissues of Tibetan pigs at follicular stage and luteal stage were enriched by GO and KEGG pathway analysis.A total of 97 GO items and78 KEGG items were enriched.In these enriched items,PI3K-Akt signaling pathway,MAPK signaling pathway,oxytocin signaling pathway and other pathways related to ovarian development or genetic information were found.Finally,real-time fluorescence quantitative PCR was used to verify 8 differentially expressed lncrnas,and it was found that the verification results were consistent with the predicted expression trend,which further confirmed the reliability of the transcriptome LncRNA data.In conclusion,this study compared LncRNA differentially expressed in ovarian of Tibetan pigs at follicular stage and luteal stage through high-throughput sequencing technology,screened target genes that could affect ovarian development through enrichment pathways related to ovarian development and genetic information,and verified LncRNA differentially expressed by real-time fluorescent quantitative PCR technology.The identified differentially expressed LncRNA affecting ovarian development and genetic information of Tibetan pigs will provide theoretical basis for genetic selection and breeding of Tibetan pigs in Tibet.
