In the process of poultry breeding and production,the quality of breeder chicken reproductive performance(such as the quality of breeder cock semen,hen egg production,sperm storage capacity,and hatching related performance of breeder eggs)has a significant impact on the production performance of chicken farms.Using effective methods to improve the reproductive efficiency of breeder chickens during the breeding and production process will provide some assistance to the breeding enterprises in their "cost reduction and efficiency increase" initiatives.In order to improve the utilization rate of breeding cocks,In this study,Liangfenghua chicken was used as the experimental object to study the chicken semen dilution technology;The study on the variation characteristics of sperm storage capacity of breeding hens provides basic data for the cultivation of high sperm storage capacity Liangfenghua chickens,and lays the foundation for reducing the frequency of artificial insemination.This study cites purchased diluents and self-designed semen diluents to control the operation and usage process,verify and compare the application effects of various diluents,in order to facilitate their practical production and application;By measuring the sperm storage capacity of Liangfenghua chickens,statistical analysis of various indicators of sperm storage capacity was conducted,and individuals with higher sperm storage capacity were selected for seed retention.The main results are summarized as follows:1.In order to explore the application effect of chicken semen diluents,this study used five diluents(WJ0,YY0,ZP1,ZP2 and ZP3)through outsourcing or self-made diluents.Through microscopic examination of the quality of diluted semen and production breeding,its actual application effect was verified.It was found that these five diluents,except ZP1 diluent,had poor fertilization effect in actual application,and the other four diluents had no significant difference from the original semen breeding,By comparing the actual application of different diluents to the stillbirth rate,it was found that the application of diluents was not directly related to the stillbirth rate.2.Phenotypic analysis was conducted on the sperm storage capacity of Liangfenghua chickens.A total of 1050 hens in three week old groups(56 week old group,58 week old group,and 60 week old group)were measured for sperm storage capacity traits.The longest fertilization days,number of fertilized eggs,effective fertilization days,maximum continuous fertilization days,and fertilization rate(fertilization rate within 21 days)were statistically analyzed,and the data showed no significant differences in various indicators among each week old group.3.Repetitive expression of sperm storage ability is an important indicator to measure whether sperm storage ability can be stably expressed in Liangfenghua chickens.By calculating various indicators of sperm storage capacity,it was found that the relevant indicators of Liangfenghua chicken’s sperm storage capacity have medium to high repeatability and stable expression ability.4.Individuals with the longest fertilization days>11 days,egg production>4(single round),and Intermittent fertilization intensity<0.2 were selected for breeding and seed retention.They were defined as the dominant sperm storage capacity group,while the other individuals were the ordinary sperm storage capacity group.It was found that the daily fertilization rate of the dominant sperm storage capacity group was significantly higher than that of the ordinary sperm storage capacity group after one artificial insemination.In addition,the average fertilization rate within 10 days in the group with superior sperm storage capacity is better than the average fertilization rate within 5 days in the entire group,indicating that genetic breeding of individuals with high sperm storage capacity can provide certain assistance in improving the reproductive efficiency of the entire chicken herd.5.After the phenotypic determination of sperm storage capacity,the body weight of each participant group was measured and divided into the dominant sperm storage group,the ordinary sperm storage group,and the test group.The average body weight of each group was compared,and no significant differences were found between the groups.This can preliminarily prove that the selection of dominant sperm storage individuals has no direct impact on poultry body weight selection.This study shows that there is no statistically significant difference in fertilization rate between self prepared semen diluents and commercial diluents,and they can be put into production and application.Compared to externally purchased diluents,self prepared diluents have a price advantage;In addition,the study on the sperm storage capacity of Liangfenghua chickens was conducted for the first time,and important parameters such as repeatability were obtained,providing basic data for the cultivation of high sperm storage capacity Liangfenghua chickens.
