| Blood physiological indicators reflect the health status of the animal body and are often used as important reference indicators for farm management,disease screening and disease diagnosis.In order to investigate the correlation between blood physiological indicators and production traits,to identify for SNPs and candidate genes associated with blood physiological indicators and to estimate the genetic parameters associated with blood physiological indicators.In this study,1422 Hu sheep(80-180 days old)were collected part of the phenotype values of production traits,and 180-dayold blood was collected to measure blood(Total red blood cells,RBC),(Haemoglobin concentration,HGB),(Red blood cell volume,HCT),(Total white blood Count,WBC),(Total platelets,PLT),and other 26 physiological indicators,and estimate their genetic parameters and Genome-wide association analysis based on whole-genome resequencing data.Gene annotation,haplotype combination,and polymorphism association analysis of significant loci were carried out through associated loci in order to obtain genetic markers associated with physiological blood indicators and provide a theoretical basis for the early selection of Hu sheep and related research on physiological blood indicators.The main results of this study are as follows:1.Correlation analysis showed that the correlation between platelet traits and red blood cell traits and white blood cell traits was weak,with a strong negative correlation between red blood cell traits and a strong positive correlation between platelet traits.The absolute values of the correlation coefficients between the various blood physiological indicators and most of the growth traits were below 0.3 and the correlations were weak.2.An analysis of the estimated genetic parameters of blood physiological indicators in hu sheep showed that the legacy of blood physiological indicators in hu sheep are traits of medium to high heritability,with heritabilities of 0.224~0.592 for red blood cell traits,Genetic correlations were high for homologous traits-0.78~0.99,low for different cell types-0.37~0.38,and low for platelet traits versus red and white blood cell traits-0.35~0.35.3.Genome-wide association analysis of 26 blood physiological indicators showed that 5 significantly associated SNPs were identified at the genome-wide level,and a total of 4 SNPs were identified for significant association with(Standard deviation of red blood cell distribution width,RDW_SD)indicators on chromosome 11(3)and chromosome 14(1);one SNP was identified for significant association with(Percentage of monocytes,MONO2)on chromosome 15.Two candidate genes related to RDW_SD(NOG and WDR87),a candidate gene related to MONO2(HBB).The results of the trait association analysis showed that the different haplotype combinations,NC_040262.1:g.60969859 C > A,NC_040265.1:g.50686485 A > C and NC_040266.1:g.51884103 C > T were significantly associated in several of the 26 indices.The different haplotype combinations were significantly associated with body weight,body height,body slope length,chest circumference and Scrotal circumference,NC_040262.1:g.60969859 C > A and NC_040266.1:g.51884103 C > T were significantly associated with body slope length,NC_040265.1:g.50686485 A > C was significantly associated with weaning weight.Different haplotype combinations and NC_040262.1:g.60969859 C > A were significantly associated with mean daily weight gain,and NC_040265.1:g.50686485 A > C and NC_040266.1:g.51884103 C > T were significantly associated with residual feed intake.In summary,based on the large-scale blood physiological index measurements and whole-genome resequencing data,this study conducted genome-wide association analysis,identified five SNPs significantly associated with blood physiological indexes in hu sheep,and annotated three key candidate genes,which can provide valuable molecular markers for molecular breeding of blood physiological indexes in Hu sheep and clues for the molecular genetic regulation mechanism of blood physiological indexes. |
