As a traditional medicinal plant,Gelsemium has anti-inflammatory,analgesic,sedation,dilatation,anti-tumor,immunity,hematopoietic function and other pharmacological properties.Koumine is the most abundant active ingredients in the China gelsemium.Koumine are used as research objects,in this study,to construct respiratory diseases caused by two different pathogens,and explore the therapeutic effect of koumine on acute lung injury,and to provide scientific basis for the clinical utilization and development of koumine in the future.1、The therapeutic effect of koumine on carrageenan-induced acute lung injury in miceThe male ICR mice were randomly divided into a model group,a koumine group(3mg·kg-1),and a dexamethasone(DEX,3 mg·kg-1)group after successful modeling.The control group was injected with normal saline without any other treatment.The mice were sacrificed 4 h after single treatment and 72 h after repeated treatment,respectively.Then the levels of inflammatory cytokines TNF-α,IL-1βand IL-6 in blood were measured,the lung coefficient and the content of MDA and MPO and the activity of SOD in lung tissue were determined.The pathological changes of lung tissue were observed.The results showed that multiple treatments with koumine could effectively inhibit the production and release of inflammatory cytokines TNF-αand IL-1β(P﹤0.05),and significantly inhibit the secretion of IL-6(P﹤0.01)in blood of ALI mice,reduce the lung coefficient(P﹤0.05),decrease the content of MPO in lung tissue(P﹤0.05),decrease the content of MDA and improving the activity of SOD(P﹤0.01),and recover the histopathological morphology of lung tissue.2、The therapeutic effect of koumine on sepsis-induced acute lung injury in miceThe sham group was treated with cecal exploration,the CLP model group,the group A(koumine,3 mg·kg-1),the group B(koumine,15 mg·kg-1),the group C(florfenicol,200 mg·kg-1),the group D(koumine,3 mg·kg-1+florfenicol,200 mg·kg-1)and the Group E(koumine,15 mg·kg-1+florfenicol,200 mg·kg-1)were all established a sepsis-induced acute lung injury by cecal ligation and puncture.Treatment was performed at 6,12,18,24and 48 h after successful modeling.then the general situation of the mice was observed,Draw a 72-hour survival curve,the wet/dry weight of lung tissue was calculated,the levels of TNF-α,IL-1β,IL-6,MPO,MDA and SOD in lung tissue of mice were determined.The results showed that koimine(15 mg·kg-1)can increase the survival rate of mice(53.33%),and the survival rate of mice(66.66%)was increased when combined with florfenicol.and inhibit the release of TNF-α,IL-1β,IL-6,MPO and MDA in lung tissue(P﹤0.01),and improve SOD activity(P﹤0.01),reduce the water content in lung tissue(P﹤0.01).Different concentrations of koumine can play different effects.However,compared with the group A,the group B may have more advantages in the improvement of symptoms and disease progression and prognosis,and the effect of combination with florfenicol is better.To sum up,koumine has significant therapeutic effect on acute lung injury induced by two different etiology in mice.The mechanism may be related to inhibiting the expression of inflammatory cytokines,improving the antioxidant capacity of the body and alleviating the degree of lung tissue damage.
