Stropharia rugosoannulata is rich in nutrients,has a wide range of environmental adaptation,strong resistance to stress and other substances,simple cultivation techniques and a broad market prospect.However,at present,the cultivation method of Stropharia rugosoannulata is still in the category of traditional agriculture,and its production efficiency is relatively low.The preliminary experiment of this research group showed that the fruiting body could be produced in the mushroom house of edible fungus factory by using rice straw cultivation,and the conditions of the fruiting body could be preliminarily explored.On the basis of the previous research,this paper optimized the cultivation substrates of the fruiting body by using the common materials(corn cob,wood chips,bran,etc.)in edible fungus factory,The effects of different C/N ratios on fruiting body yield,fruiting body time,fruiting body rate and fruiting body characters were further studied.The changes of enzymes related to carbon and nitrogen metabolism under different C/N ratios were preliminarily explored to optimize the industrial production process parameters of Stropharia rugosoannulata.The contents and results of the study are as follows:(1)Huqiu No.1(CGMCC5.2211),which was provided by Institute of edible fungi,Shanghai Academy of Agricultural Sciences,was selected as the research object.The results showed that different C/N formulations had different effects on the growth of mycelia and the higher C/N ratio was,the higher growth rate of mycelia was.The fruiting bodies grown on the containers with square baskets were more thick and thicker than those directly covered with soil.The fruiting bodies were brighter in color and fuller in color,but the yield was not much different from that of the containers.Further study showed that there were significant differences in the fruiting body yield among different C/N ratios,and the yield of C/N 60 was the highest.The growth of mycelium and fruiting body of Stropharia rugosoannulata could be affected by the different ratio of carbon to nitrogen of cultivation materials.Further experiments showed that the contamination rate of each formula was reduced in the fruiting body growth stage,the fungus with C/N ratio of 40 was completely polluted,while the fungus with C/N ratio of 88(pure corn cob)did not produce fruiting body.The fruiting body yield of the C/N ratio of 50 formula with the second highest pollution rate was 65.5%,while the fruiting body yield of the C/N ratio of 70 formula with the lowest pollution rate was only 18.5%.The formula with C/N ratio of 60 had the earliest primordium,moderate contamination rate,moderate fruiting body yield and the highest yield.We preliminarily selected the optimal formula of C/N ratio of60 for the cultivation of Stropharia rugosoannulata.(2)The activities ofβ-1,3-GA,β-GC,Cx,GS and GOGAT of formula 60,which formed the earliest primordium,were maintained at a high level during the mycelial growth stage of Stropharia rugosoannulata.However,the activity of these enzymes in the uncultured formula 88 was always at a very low level.In the process of growth and development,the activities of enzymes related to carbon and nitrogen metabolism were high,which improved the utilization of carbon and nitrogen,provided a better material and energy basis for the further growth and development of the fruiting body,and promoted the earlier reproductive growth stage of the fruiting body,so as to achieve the effect of increasing the fruiting body yield.(3)It was found that the formation time of primordium in the second mode was4 days earlier than that in the first mode,by improving the first mode and reducing the relative humidity of the air in the early stage of culture,we can reduce the diseases and insect pests of bacterial packets.Further study showed that the humidity in the early stage of fruiting body formation should not be too high,and appropriate adjustment should be made.In this experiment,the concentration of CO2was between700-1500ppm,and the variation trend of CO2content was similar to that of air temperature,which could reflect the metabolic situation of the mycelium of Stropharia rugosoannulata in the fruition stage from the side.Temperature and humidity are important factors affecting the growth and development of Stropharia rugosoannulata.Different conditions will produce different results,compared with the second mode is better.To sum up,the optimal C/N ratio of 60 was selected and the factory production parameters were optimized in this study.It can produce the fruiting body of mushroom with high yield and good quality,shorten the cultivation period and improve the production efficiency.The results revealed the effects of the changes in the activities of enzymes related to carbon and nitrogen metabolism on the growth and development of Stropharia rugosoannulata.On the one hand,this study provided an experimental basis for studying the mechanism of carbon and nitrogen regulation on the fruiting body development of Stropharia rugosoannulata,and also laid a practical foundation for future research on the industrial production of Stropharia rugosoannulata.
