| Pholiota nameko,an edible and medicinal fungus of the genus Pholiota,also known as sliding mushroom,bare cap yellow umbrella,pearl mushroom.It is now widely cultivated for food and traditional medicine in China and Japan.Pholiota nameko is widely cultivated and has become a common edible mushroom with a sticky and slippery nature of its cap,a more typical wood-rotting fungus with various biological activities such as anti-inflammatory,anti-hyperlipidemic and anti-tumor activities.With the expansion of the sliding mushroom industry,the by-products(Pholiota nameko eluting mucilage)produced are also increasing,and these by-products are often not applied with maximum efficiency and have a polluting effect on the environment.To address this problem,this thesis rationalizes the use of polysaccharides,proteins,nucleic acids and other components in the Pholiota nameko eluted mucus to turn waste into treasure and reduce pollution while rationalizing the use of eluted mucus to prepare wound dressing and investigate its effect on wound healing.The main research results are as follows:1.Preparation and composition characterization of Pholiota nameko eluted mucus dressing:Pholiota nameko was used as raw material,and the mucus on the surface of Pholiota nameko was washed away by elution,and the mucus was lyophilized and dried to obtain Pholiota nameko eluted mucus wound dressing.The total sugar content of the lyophilized and dried dressings was 43.2%and 28.9%,respectively.The results of detecting their monosaccharide composition,amino acid types and contents showed that except for arginine,phenylalanine and lysine,the contents of other amino acids in the dried powder were higher than those in the lyophilized powder;additional rhamnose was detected in the dried dressing,and additional galacturonic acid was detected in the lyophilized dressing.2.Evaluation of antioxidant activity of Pholiota nameko eluting mucilage dressing:the antioxidant activity of Pholiota nameko eluting mucilage was evaluated by DPPH,hydrogen peroxide,superoxide anion and hydroxyl radicals,respectively,and the results showed that at 5mg/m L,the scavenging rates of freeze-dried and dried dressings were 46.50%and 43.75%for DPPH,and the scavenging rates of superoxide anion radicals were 71.68%and 68.42%,and the scavenging efficiency of hydroxyl radicals was 54.63%and 51.22%,respectively,while the highest scavenging efficiency of H2O2 was achieved at 1 mg/m L,92.75%and 91.75%,respectively.3.Assessment of pro-cell proliferation and migration ability of Pholiota nameko eluting mucilage dressing:the proliferation ability of lyophilized and dried dressings on mouse fibroblasts(NIH/3T3)was assessed by cell viability and Ed U proliferation assays,which showed that both could promote cell proliferation with proliferation rates of 156.65%and146.18%,respectively.The cell scratch assay assessed the migration rate of NIH/3T3 cells after treatment with lyophilized and dried dressings as and 156.2%and 144.73%,respectively.4.Assessment of wound healing capacity of Pholiota nameko eluting mucus dressing:the wound healing capacity of Pholiota nameko eluting mucus was assessed using a mouse skin wound model.The experimental results showed that the expression levels of inflammatory factors TNF-αand IL-6 at the trauma site of mice were reduced at 3 days.After the treatment,HE staining,MASSON staining and CD31 fluorescence image analysis showed that mushroom eluting mucus could promote skin wound healing by promoting the regeneration of hair follicle appendages and capillaries under the skin wound,and the remodeling of granulation tissue and collagen tissue.In conclusion,Pholiota nameko eluting mucilage can accelerate wound healing by promoting cell proliferation and migration and reducing inflammatory response. |
