Tibetan pig is a small pig breed with adaptability to high altitude hypoxia environment in China.Studying the adaptation mechanism of Tibetan pigs to hypoxic environment is helpful to understand and explain various plateau physiological phenomena,which can provide new strategies for the treatment of related diseases.There have been many reports on the hypoxia adaptability of Tibetan pig heart and lung,but there are relatively few studies on other oxygen-consuming organs such as muscle and kidney.As a complete individual,each tissue and organ will not only play its own unique role in dealing with hypoxic environment,but also coordinate with each other.Therefore,it is equally important to study the adaptation and function of Tibetan pig muscle and kidney to hypoxic environment.In this study,the RNA-seq data of kidney and muscle tissues of Tibetan pigs and Banna small-ear pigs were analyzed to explore the mechanism of hypoxia adaptability of Tibetan pigs.1.By comparing the transcriptome sequencing data of Tibetan pigs and Banna small-ear pigs,there were 1928 differentially expressed genes in the main effect group of kidney varieties and 1949 in muscles.There were 64 differentially expressed genes in the main effect group of kidney altitude and 817 in muscle.There were 3570 differentially expressed genes in the kidney interaction group and 1229 in the muscle.There were 1135 differentially expressed genes in the kidney variety effect plus interaction group and 123 in the muscle.There were 4099 differentially expressed genes in the kidney elevation effect plus interaction group and 184 in the muscle.2.The differentially expressed genes VCAM1,ITK,VEGF,VEGFA,FLT1,ACVRL1,EPO and ENO1 related to hypoxia adaptation of Tibetan pigs were screened.These genes play an important role in the kidney adaptation of Tibetan pigs to hypoxic environment by participating in immune,angiogenesis,erythropoiesis and glucose metabolism.3.Through co-expression network analysis,genes HILPDA,PNPLA2,LYRM1,UBE2 B,RSPO3 and MCUB specifically expressed in muscle were screened.They play an important role in Tibetan pig muscle tissue adaptation to hypoxic environment by regulating glucose metabolism and lipid metabolism.In this study,the transcriptome data of Tibetan pig kidney and longissimus dorsi muscle were analyzed by two-factor differential expression analysis of altitude and variety and the regulatory network of co-expressed gene clusters.The differentially expressed genes that play an important role in hypoxia adaptation of Tibetan pig kidney were screened : VCAM1,ITK,VEGF,VEGFA,FLT1,ACVRL1,EPO,ENO1,PGK1 and tissue-specific expression genes HILPDA,PNPLA2,LYRM1,UBE2 B,RSPO3,MCUB in Tibetan pig muscle hypoxia adaptation.
