Asparagus bean [Vigna unguiculata(L.)ssp.sesquipedalis Verd.] is a worldwide legume crop.Because its tender pods are rich in nutrients,they are popular with consumers.With people paying more attention to the nutritional quality of vegetables,there is an urgent need for high-quality asparagus bean varieties to be marketed.Previous researchers have carried out beneficial research on the nutritional quality traits and inheritance of asparagus bean,but the varieties and trait indicators used are few,which is difficult to reflect the overall level of nutritional quality of asparagus bean population.In order to improve the understanding of the nutritional quality traits of asparagus bean and promote the genetic improvement of quality,this study was based on the 29 quality traits data of the pod systematically measured by 243 asparagus bean varieties split plot(variety+year)test design method,revealed the characteristics and distribution characteristics of the trait data,estimated the genetic parameters,and carried out comprehensive quality evaluation and germplasm screening;Combined with phenotypic data and genome re-sequence data,GWAS analysis was carried out to excavate significant loci and candidate genes related to seven quality traits including valine,isoleucine,methionine,water-soluble protein,flavonoid and copper content,providing a basis for genetic analysis of quality traits,functional gene validation and quality trait improvement of asparagus bean.The main findings are as follows:(1)This paper uses the B/S structure,My SQL as the database,and PHP language to compile,standardize and input 243 asparagus bean germplasm information,and build the asparagus bean germplasm resource quality character database,which has the function of resource information query and statistical analysis.(2)The results of field experiment and biochemical analysis showed that the variation coefficient of 29 quality traits of asparagus bean ranged from 10.34% to 66.41%;The difference of most characters between varieties is very significant,and there are also differences between years;Based on the characteristic data,the main nutrient composition table of asparagus bean was drawn and linked with the quality character database;The data characteristics and distribution characteristics of asparagus bean quality traits were displayed in the form of violin distribution map;The generalized heritability of 16 traits,such as vitamin C and dry matter content,is more than 80%,and the generalized heritability of leucine and methionine content is less than 40%;There are 98 correlation coefficients between the two amphoteric traits that reach the extremely significant correlation level,and the correlation between amino acid components is strong;The cluster analysis can divide 243 varieties into five groups,42% of which are clustered in group II,representing the benchmark level of asparagus bean.Groups I,III and V show high sugar,amino acid and protein content respectively,and individual varieties with extreme characteristics are scattered in group IV;The principal component analysis transformed all the characters into 9 principal components,with the cumulative contribution rate of 65%.According to the comprehensive evaluation score,25 excellent germplasms such as Anthurium bicolor and Ejiang No.1 were selected.(3)10000 high-quality SNP markers with polymorphism were screened using the genome re-sequence data of asparagus bean varieties,and the whole-genome association analysis was performed with the phenotypic data of quality traits of corresponding varieties.173 significant SNP loci were detected for texture and seven quality traits including valine,isoleucine,methionine,water-soluble protein,flavonoid and copper content.According to the existing annotation information of asparagus bean genome,focusing on the biological process and molecular function of genes involved,referring to the genome of Arabidopsis thaliana,a model crop,a total of 108 candidate genes related to 7 personality traits were screened by searching at 100 kb on the left and right sides of the significant SNP sites.
