| Lilium is one of the most important commercial flowers in the world,with highly ornamental and economic value and a long cultivation history.The lily cultivars in China’s market are heavily dependent on imports,so it is particularly important to have excellent new varieties of lily with independent property rights.Lily breeding methods are diverse,and polyploid breeding is one of the important methods for breeding new varieties.This method can help to obtain new ploidy lilies potentially restore the fertility of triploid lily.In this study,the scales of allotriploid 1712 lily(OTO)were treated with different concentrations of Oryzaline and different time.The main research results are as follows:1.Using 1712 tissue culture seedling scales as materials,soaking with 0.002%Oryzaline for 8 was the best treatment for chromosome doubling,with a doubling rate of63.3%.2.The GISH results showed that the total number of chromosomes of 1712 were 36,including 23 O-chromosomes 23,11 T-chromosomes and 2 OT-recombinant chromosomes.After successful chromosome doubling,the total numbers chromosomes become 72,consisting of 46 O-chromosomes,22 T-chromosomes and 4 OT-recombinant chromosomes.3.Compared with 1712,the obtained hexaploid lily tissue culture plantlets had thicker leaves,shorter and wider leaf shapes,and some leaves were deformed;Their scales thicken and clumped to grow;Root system becomes thicker and fewer;Growth were slow.After transplantation,the leaf surface of the plant becomes unsmooth,has more small folds,and grows slightly slower.The results showed that the morphological differences of tissue culture seedlings and transplanted plants were very different between the two. |
