Enterocytozoon bieneusi(E.bieneusi),Giardia lamblia(G.lamblia)and Cryptosporidium(Crypto)are three important intestinal parasites,which can infect human beings,domestic animals and pets.Dogs,as one of the animals that have the closest contact with human beings,are also important hosts of these three intestinal parasites.In order to understand the infection of three kinds of parasites in canine intestine of some areas of Harbin and analyze the risk of zoonosis,the epidemiological investigation was carried out.At the same time,the genotypes or species of the three intestinal parasites were identified.In some areas of Harbin,408 fresh dog feces were collected,including 98 pet hospitals,74 in the community,139 stray dog shelters,97 pet markets.Nested PCR and sequencing were used to detect the molecular biology and genotype of E.bieneusi,G.lamblia and Crypto.PCR results showed that 26 E.bieneusi positive samples were detected and the infection rate was 6.4%.The results showed that the infection rate of stray dog shelters was 9.4%,7.2% in pet market,3.1% in pet hospital and 4.1% in community,and the infection rate was significantly different in different places.Based on the ITS sequence of E.bieneusi,26 positive samples were genotyped.A total of 8genotypes were identified,including 4 known genotypes,Pt Eb IX(n = 15),Ebp C(n = 2),D(n = 2),WW8(n = 1),with Pt Eb IX as the dominant genotype and 4 unknown genotypes,temporarily named DL132(n = 3),DL92(n = 1),DW33(n = 1)and XF17(n = 1).The infection rate was 3% in 12 G.lamblia positive samples.The infection rate in different groups was 4.3%,4.1%,2.6% and 2% respectively in pet market and pet hospital,and no positive samples were found in Nangang district.Based on the BG gene sequence of G.lamblia,12 positive samples were genotyped.A total of 2 G.lamblia aggregates were identified by sequence analysis,including aggregates C(n = 4)and D(n = 8).Crypto was detected in 7 positive samples with infection rate of 1.7%.The results of different groups showed that the infection rate of stray dog shelters in the source group was the most serious,with an infection rate of 3.5%,followed by the pet market,with an infection rate of 2.1%.No positive samples of Crypto were found in pet hospitals and communities,and there was a significant difference in the infection situation between different source areas.The infection rate of each group in the gender group was 1.8% for males and 1.7% for females,with no significant difference.Based on the SSUr RNA gene sequence of Crypto,7 positive samples were genotyped.Two genotypes,C.canis(n = 4)and C.parvum(n = 2),were identified by sequence analysis.E.bieneusi genotypes D and Ebp C belonging to Group1 and C.canis genotypes and C.parvum detected in this study were both zoonotic,so this study showed that E.bieneusi and Crypto have higher risk of zoonosis.
