Observation On The Therapeutic Effect Of GC376,GS-441524 And Feline Interferon-omega On Feline Infectious Peritonitis

Abstract/Summary:
Feline infectious peritonitis(FIP)is a chronic,systemic,and lethal disease caused by the Feline infectious peritonitis virus(FIPV),which lacks effective means of prevention due to the specificity of its pathogen.It is a serious threat to the life and health of felines and causes many problems for animal owners.The existing treatment methods in China are basically symptomatic,and there is no targeted treatment drug.In recent years,foreign countries have developed corresponding drugs,mainly GC376,GS-441524,and Feline Interferon-omega(Fe IFN-ω),which have achieved good therapeutic effects in experimental studies,but there are no reports on the therapeutic effects of the combination of GS-441524 and Fe IFN-ω.The above three drugs have not been introduced into China yet,but biologics of the drugs have been granted by scientific institutions in China in recent years,but systematic clinical validation has not been carried out.A total of 156 felines with naturally infected FIP were admitted to a chain animal hospital in Beijing from January 2021 to December 2022.The age,sex,breed,body condition score,type of disease,clinical symptoms,radiographic examination results,and complications of the felines were counted,and the correlation between the above factors and the occurrence of FIP was analyzed to summarize the development pattern of FIP.The 156 felines with FIP were divided into three groups and treated as follows,GC376 treatment group(group 376): 31 felines,subcutaneous injection of GC376 15 mg/kg once/12 h;GS-441524 treatment group(group 441): 98 felines,subcutaneous injection of GS-441524 7.5 mg/kg once/24 h;GS-441524 and Fe IFN-ω combination treatment group(441+ω group): 27 felines,subcutaneously injected with GS-441524 7.5 mg/kg once/24 h and subcutaneously injected with GS-441524 at the same time as Fe IFN-ω 400,000 IU/kg once/24 h in the first week of treatment initiation;all groups took appropriate symptomatic treatment according to the condition of the affected felines,and continued to administer the drug for 8 w.The results of blood cell sorting count(white blood cell(WBC),lymphocyte(LYM),lymphocyte percentage(LYM%),red blood cell(RBC),hemoglobin(HGB),and hematocrit(HCT)),blood biochemical indexes(total protein(TP),albumin(ALB),globulin(GLOB),A:G,amylase(AMY),and total bilirubin(TBIL)),and serum amyloid A(SAA)were recorded before treatment in each group of felines.The above indexes were followed up and monitored,and the changes in blood indexes before and after treatment in each group of felines were compared,and the effects of drugs on the above blood indexes in each group were analyzed.The clinical status and blood indexes of the felines in each group during the treatment were tracked and monitored,the efficiency of the drugs in each group was statistically analyzed,the regression of the felines in each group and the treatment period were counted,and the treatment effects of GC376,GS-441524,and GS-441524 in combination with Fe IFN-ω were compared.Of the 156 FIP-affected felines participating in this trial,70.51% were young felines and 29.49%were adult felines.The consultation rate of male felines was much higher than that of female felines,with a male-to-female ratio of 2:1.54.49% of the introduced breed felines were slightly higher than those of Chinese field felines(45.51%).The incidence of the exudative form was 62.82%,which was higher than that of the non-exudative form(37.18%).Most of the affected felines had a body condition score of 5 or less at the first visit(80.13%).Clinical signs were dominated by loss of appetite(64.74%),depression(57.69%),fever(39.74%),and weight loss(37.18%).The most frequent complications were hepatobiliary disease(32.69%)and simple hyperbilirubinemia(27.56%).After 8 weeks of drug treatment,the clinical symptoms of FIP felines all improved significantly.TP and TBIL in the 376 group were significantly reduced,and LYM,RBC,and ALB were significantly increased;WBC,TP,GLOB,AMY,and SAA in the 441 group were significantly reduced,and LYM,RBC,HGB,HCT,and A:G were significantly increased;TP,GLOB,AMY,and SAA in the 441+ω group were significantly reduced,and RBC,HGB,HCT,and A:G were significantly increased.The efficiency of the 441+ω group was higher than that of the 376 group,with a significant difference(P < 0.05).The survival rate of the felines in each group was not significantly different.The duration of medication in the 441 group was shorter than that of the 376 group,with a highly significant difference(P < 0.01);the duration of medication in the 441+ω group was shorter than that of the 376 group,with a highly significant difference(P < 0.01);the dosing time of the 441+ω group was shorter than that of the 441 group,and the difference was significant(P < 0.05).In conclusion,the therapeutic effect of GS-441524 was better than that of GC376;the therapeutic effect of the combination of GS-441524 and Fe IFN-ω was better than that of GS-441524 alone.It provides a dosing reference and theoretical support for the domestic clinical treatment of FIP,and provides a basis for the introduction and application of the above clinical trial drugs.
