Wheat(T riticum aes tivum L.)is a widely grown grain crop in the world,which cultivation area and production are among the world’s top.However,wheat is often subje cted to abiotic stress es such as drought,high temperature and high salinity during growth,resulting in delayed growth an d reduc ed yield.As a class of transcri ption f actors commo nly found in plants,C 2H2 zinc finger proteins play an important role in the r egulation of plant growth and stress response.Currently,studies on the regulat ion of C 2H2-type tran scription factors in abiotic stresses m ainly focus on Arabidopsis and ric e,while the function of C2H2-type transc ription factors in whea t is rarely r eported.In this study,T a Z F P 2 3,a member of C 2H 2transcription factor family in whe at,was selected to explore its function in regulating abiotic stress response.The mai n results are as follows:1.Bioinformatic analysis showed that whe at TaZFP23 coding sequence w as720 bp in length,en coding 238 amino a cid residues.Ta ZFP 23 was a typic al C2H2zinc finger prot ein with two C 2H2zinc finger domain,on e EAR motif and none transmembran e domain.The resul ts of promoter cis-acting element analysis showed that TaZFP23 might be involved in abiotic stre ss response and phytohormone signaling.2.The full-length se quence of whe at TaZFP23 was success f u lly cloned,and the expression v ector p CAMBIA2300 with GFP label was constructed.The result of subcellular location showed that TaZFP23 encoded a nuclear protein.The result of trans cri ptional a ctivity identification suggested t hat Ta Z FP 23 did not possess transc riptional activation acti vity.3.The wh eat TaZFP23 was tr ansfor med into wild-type Arabidopsis t h al i a n a.Two homozygous A r a b i d o p si s lines overexpressing T a Z F P 2 3(OE-1,OE-2)we re obtained for further functional resea rch through anti biotic scr eening,and DNA and prot ein level identification.4.The gemination rate of wild-typ e Arabidopsis and TaZFP23overexpr essing Arabidopsis was calcul ated under salt,mann itol,and ABA treatment,and the r esults showed that the gemination rate of Arabidopsis overexp r essing TaZFP23 was lower than that of wild-type seed s.Additionally,the survival r ate o f Arabidopsis over exp ressing TaZFP23 wa s also lower than that of wild-type.P hysiological and biochemical pa ramete rs showed that th e antioxidant enzyme activity and pr oline content of Arabidopsis leaves overexpr essing TaZFP23 wer e lower tha n those of wild-type under salt and drought stress conditions,while the MDA content was higher than that of wild-type.q RT-PCR resu lts showed that the expression levels of several stress-related mark er gen es in Arabidopsis over expressing TaZFP23 were low er than those of wile-type un der drought and salt stress conditions.
