| Ginger is a asexual reproduction crop,which is prone to seed degeneration in the production process.In recent years,tissue culture virus-free has been widely used in production.However,the management of test tube seedling transplantation is more complex,so it is important to induce the root and stem of ginger tissue culture seedlings to expand and produce miniature ginger,which is important for the production of non-toxic ginger seedlings.Therefore,this article conducted a study on the induction of swelling of micro ginger in ginger tissue culture seedlings.The main results are as follows:1.In ginger tissue culture seedlings,sucrose concentration plays an important role in the induction of micro ginger.The results of this experiment indicate that when the sucrose concentration is below 140 g·L-1,the fresh weight of micro ginger increases with the increase of sucrose concentration.Within the range of sucrose concentration of 60-115 g·L-1,the swelling of micro ginger can be clearly observed,and the higher the sucrose concentration,the more obvious the effect.When the concentration of sucrose is below 60g·L-1,the induction effect on micro ginger is very weak,and the swollen parts are basically not observed from the phenotype.When the concentration of sucrose is higher than 115 g·L-1,the effect of inducing the swelling of micro ginger begins to weaken.Excessive sucrose concentration will inhibit the growth of ginger tissue culture seedlings.Sucrose concentration can also affect the growth of ginger tissue culture seedlings’roots,stems,and leaves.In this experiment,a sucrose concentration of 60 g·L-1 was the most favorable for the growth of roots,stems,and leaves.2.The level of mineral elements in MS medium has a significant impact on the growth of in vitro seedlings.The experimental results showed that appropriately increasing the level of mineral elements had a promoting effect on the growth of in vitro seedlings,but the effect on the induction of micro ginger was not as significant as that brought by sucrose concentration.In summary,when the concentration of sucrose in the culture medium is 90 g·L-1 and the level of mineral elements is 1 MS,it will not have a significant impact on the growth of ginger tissue culture seedlings,and it is more significant in inducing micro ginger.3.Light quality also has a significant impact on the growth of ginger tissue cultured seedlings.The experiment used four treatments to explore the induction effect of W(white light),W+G(white light+green light),W+R(white light+red light),and W+B(white light+blue light)on micro ginger in ginger tissue culture seedlings.Under the same light intensity,the induction effect of four different light quality combinations on micro ginger was different.For the fresh weight of micro ginger in whole ginger tissue culture seedlings,the W+G combination treatment showed the best result,significantly higher than other treatments.Different light quality combinations have different effects on various growth indicators.Overall,the W+G combination treatment has a greater impact on the growth of ginger tissue cultured seedlings,followed by the W+R combination treatment.4.From the perspective of the effect of light quality on ginger tissue culture seedlings,the combination of W+G treatment had the best induction effect on miniature ginger.Therefore,W+G was selected for metabonomic analysis and endogenous hormone determination.78differential metabolites were selected.According to the primary classification of substances,there are mainly 10 categories,including amino acids and their derivatives(16),flavonoids(20),lipids(10),terpenes(5),organic acids(3),alkaloids(4),phenolic acids(6),nucleotides and their derivatives(2),lignin and coumarins(1),and other categories(11).A total of 24differential metabolites were allocated to these 33 metabolic pathways,of which 15 were involved in the metabolic pathway,accounting for 62.5%of the total,followed by 8 differential metabolites involved in the secondary metabolic pathway,accounting for 33.3%of the total. |
