The Huaxi District of Guiyang City is a typical karst geological area.Against this background,the unique characteristics of low latitude,high altitude,and low sunlight in the area have led to the rapid development of the tea industry in the area.As an important agricultural pest that has a negative impact on the quality and yield of tea,Empoasca onukii,the"3R"problem caused by long-term application of chemical pesticides,not only endangers human health but also affects foreign trade exports,seriously hindering the healthy and sustainable development of the tea industry in the region.It is necessary to find efficient and low toxicity prevention and control methods to replace traditional chemical pesticide prevention and control.This paper first analyzed the relationship between the occurrence dynamics of Empoasca onukii and the meteorological factors,and finally compared the fumigation toxicity of the two essential oils,and identified and analyzed the detoxification gene of Empoasca onukii’s response to patchouli essential oil,in order to provide a theoretical basis for the appropriate prevention and control and further development of efficient natural pesticides,and promote the healthy development of the tea industry.The main results of this article are as follows:1.There is a close correlation between the occurrence dynamics of Empoasca onukii and meteorological factors in Zhaosi Tea Garden,Huaxi District,Guiyang City.It was found that the occurrence dynamics of Empoasca onukii are bimodal,with two peak periods in late July and early September,respectively.Stepwise regression analysis,partial correlation analysis,and path analysis indicate that the occurrence dynamics of Empoasca onukii are closely related to two meteorological factors:monthly average maximum temperature(X6)and precipitation(X2).They are the main factors affecting the occurrence dynamics of Empoasca onukii.The tea garden ecosystem,consisting of various plants that are suitable for karst growth,also has a certain impact on the occurrence of Empoasca onukii.2.Discovered that Patchouli alcohol,δ-guaiacumene andα-guaiacumene are the top three components in the essential oil content of patchouli(61.61%);Camphor,safrole,and eucalyptol are the top three components in cinnamomum camphora essential oil content(50.68%).The LC50 values of Empoasca onukii treated with two essential oils for 36 h are0.474μL·m L-1 and 1.204μL·m L-1,respectively.Both plant essential oils have the potential to control Empoasca onukii,but further experimental analysis is needed on the detailed components that affect Empoasca onukii.3.The transcriptome sequencing results of patchouli essential oil stressed Empoasca onukii showed that a total of 2309 differential genes were found in the control group and the treatment group,including 1597up-regulated genes and 712 down regulated genes.The differential genes were identified and found to involve many detoxification gene families(P450 gene family,GST gene family,Car E gene family,ABC gene family,UGT gene family),and these gene families were analyzed by bioinformatics.It suggested that these genes may play an important role in the essential oil stress on Empoasca onukii.We also found genes(SOD genes,CAT genes,POD genes),HSPs gene and OBPs gene of redox related gene families.
