Schistosomiasis japonicus is a zoonotic infection caused by Schistosoma japonicum that is predominantly endemic in developing countries and remote areas,infecting up to 258 million people,and is of significant medical and economic importance in endemic countries and regions.There is no effective vaccine for Schistosoma japonicum on the market,and chemotherapy with praziquantel is the main strategy to control schistosomiasis.However,reliance on and widespread use of PZQ can reduce the susceptibility of Schistosoma japonicum to it and increase the risk of drug resistance.Therefore,there is a need to find a vaccine or drug that can interrupt the transmission of Schistosoma japonicum and is effective against the reproduction of Schistosoma japonicum.In this study,we extracted the active ingredient SLs from the rhizomes of Ferulic Sesquiterpene Lactones against Schistosoma japonicum and observed its killing effect on Schistosoma japonicum.This study was divided into 3 main aspects as follows.1.To observe the killing effect of Ferulic Sesquiterpene Lactones on both juvenile and adult worms of Schistosoma japonicum in vitro.Mice were infected with Schistosoma japonicum caecal larvae for 18 d,and the adult worms were collected by hepatic portal vein perfusion method,35 d after infection.The adult worms were incubated in DMEM culture medium,and different concentrations of ferulic sesquiterpene lactones and praziquantel solutions were added to the culture medium for 3 days.The results showed that the different doses of ferulic sesquiterpene lactones were effective in killing adult worms,and the survival rate and viability reduction rate of adult worms were highly significant(p<0.01)in the high dose group and the blank control group,and not significant(p>0.05)in the praziquantel reagent group;the high dose ferulic sesquiterpene lactone group was effective in killing child worms,and the survival rate and viability reduction rate of child worms were different from the control group.The differences between the high-dose ferulic sesquiterpene lactones and the control group were significant(p<0.05)and not significant(p>0.05).The above results indicate that ferulic sesquiterpene lactones have the effect of antibodies against female,male and child Schistosoma japonicum.2.To observe the killing effect of Ferulic Sesquiterpene Lactones on both juvenile and adult worms of Schistosoma japonicum in vivo.The mice were administered on the 18 th and 35 th d after infection.The experimental groups were divided into praziquantel group and ferulic sesquiterpene lactone group,and a blank control group was set up.The mice were dissected and killed after 7w of administration,and the number of worms was collected from the hepatic portal vein to calculate the rate of worm reduction and female reduction.The liver was digested to calculate the rate of egg reduction.The results showed that the different doses of ferulic sesquiterpene lactone reagent were effective in killing adult worms.The difference between the reduction rate and egg reduction rate of the high-dose group and the blank control group was highly significant(p<0.01),and the difference with praziquantel reagent was not significant(p>0.05);the killing effect of the high-dose ferulic sesquiterpene lactone group on child worms was significantly different from that of the blank control group(p<0.05),and the difference with praziquantel reagent was not significant(p>0.05).The above results showed that ferulic sesquiterpene lactones were effective in killing the adult and juvenile stages of Schistosoma japonicum cultured in mice,and the higher dose of ferulic sesquiterpene lactone reagent was more effective in killing the adult stage at different developmental stages.3.Effect of Ferulic Sesquiterpene Lactones on the immune regulation of Schistosoma japonicum-infected mice immune regulation in mice infected with Schistosoma japonicum The liver tissues were sectioned at 10 w,12w,14 w and 16 w after the administration of the drug to mice 18 days after infection with Schistosoma japonicum caecilians,and the changes in the histopathology of liver granulomas in Schistosoma japonicum-infected larvae were observed under the microscope by ferulic sesquiterpene lactones.The results showed that ferulic sesquiterpene lactones inhibited the inflammatory response of granulomas,reduced the degree of fibrosis caused by Schistosoma japonicum in the liver and reduced the damage to the liver.
