In this study,22 Cyperus esculentus resources from different strains were used as materials.Under the condition of salt stress in saline alkali land of the Yellow River Delta,through correlation analysis,Principal Component Analysis and subordinate function comprehensive evaluation,salt-tolerant varieties in different growth stages of Cyperus esculentus were selected,clear salt-tolerant screening index of each period for Cyperus esculentus.The results provide reference for salt tolerance identification of Cyperus esculentus in different stages.SRAP molecular marker technology was used to explore the genetic diversity and reveal the genetic relationship among the germplasm resources of Cyperus esculentus,providing theoretical basis for the application of high-quality germplasm resources of Cyperus esculentus and breeding of new varieties.Main research results:1.Under the condition of salt stress,10 growth and physiological indexes of 22 Cyperus esculentus materials were investigated at seedling stage,among which the soluble protein content of different Cyperus esculentus strains had the greatest difference.According to the membership function value and salt tolerance evaluation criteria,1 high salt tolerant material,3 salt tolerant material,10 medium salt tolerant material and 8 salt sensitive material were screened out.The salt tolerance indexes of Cyperus esculentus germplasm resources at seedling stage were tillering number,soluble protein content,PRO,MDA,SOD and CAT.2.Under salt stress,9 growth and physiological indexes of 22 Cyperus esculentus materials were investigated at tillering stage,among which POD of different Cyperus esculentus materials was the most different.According to the membership function value and salt tolerance evaluation criteria,3 high salt tolerant material,3 salt tolerant material,15 medium salt tolerant material and 1 salt sensitive material were screened out.The salt tolerance indexes oⅡf Cyperus esculentus germplasm resources at tillering stage were plant height,PRO,POD,SOD and CAT.3.Under the condition of salt stress,six growth and physiological indexes of 22 Cyperus esculentus materials were investigated at the harvest stage,and the difference of dry weight per hundred grains of different strains of Cyperus esculentus was the greatest.According to the membership function value and salt tolerance evaluation criteria,3 high salt tolerant material,11 salt tolerant material,6 medium salt tolerant material and 2 salt sensitive material were screened out.The salt tolerance indexes of Cyperus esculentus germplasm resources at the harvest stage were plant height,number of single hole,dry weight of single hole and yield.4.The optimal SRAP-PCR system was established by single factor test and orthogonal test.The primer was 0.4 μmol/L,the mixed enzyme volume was 8.5 μmol/L and the template DNA content was 40 ng.54 primer combinations were selected from 153 primer combinations for genetic diversity analysis.A total of 495 polymorphic loci were amplified from 22 Cyperus esculentus materials,and the average polymorphism rate was 90.29%,the genetic consistency range was 0.96 ~ 0.55,the genetic distance range was 0.45 ~ 0.04.The 22 Cyperus esculentus germplasm resources were divided into five groups at the genetic similarity coefficient of 0.83,and the first group was divided into six subclasses at the genetic similarity coefficient of 0.91.Among them,"Cyperus esculentus 1","Cyperus esculentus 9" and "Cyperus esculentus 11" became a subclass respectively,and other materials were distributed in clusters.
