In this study,‘Hutai.8’ ‘Red Globe’ and ‘Suxin 1’ were used as scions,‘5BB’ ‘1103P’ and ‘Shanhe 1’as rootstocks,greenwood grafting and hardwood grafting were carried out.By investigating the grafting survival rate of different stock-scion combinations,the stem diameter,shoot length and peroxidase activity of different stock-scion combinations were compared,and the grafting compatibility of different stock-scion combinations was comprehensively evaluated.It provides a theoretical basis for the early grafting compatibility index of different stock-scion combinations and the screening of excellent stock-scion combinations.The main research results are as follows :(1)By comparing the survival rate of greenwood grafting and hardwood grafting of different scion combinations,it was found that the grafting survival rate of ‘Suxin 1’ / ‘Shanhe 1’ was the highest,reaching more than 70.00%.In addition to the combination ‘Red Earth’ / ‘Shanhe No.1’ and ‘Suxin No.1’/ ‘5BB’,the survival rate of hard branch grafting was lower than that of greenwood grafting.(2)After greenwood grafting,the peroxidase activity,soluble protein and soluble sugar content in the graft union of stock-scion were significantly positively correlated with the grafting survival rate,and the correlation coefficients were 0.913,0.938 and 0.837,respectively.However,the superoxide dismutase activity was significantly negatively correlated with the grafting survival rate,and the correlation coefficient was-0.909.(3)It was found by observing anatomical structure of stock-scion graft union of greenwood grafting,and it was found that the callus and parenchyma cells at the junction of the scion with high grafting survival rate were easy to form,and the isolation layer was easy to disappear,forming a new vascular bundle;but the grafting survival rate of low combination of scion joint is not close,there is obvious isolation layer,there are few new parenchyma cells.(4)After hardwood grafting,the shoot length,shoot diameter,root number,maximum root length,maximum root diameter,root activity,soluble sugar and soluble protein content of root system,superoxide dismutase activity of root system of each stock-scion combination were extremely significantly positively correlated with grafting survival rate and the correlation coefficients were all greater than 0.910,while peroxidase activity was extremely significantly negatively correlated with grafting survival rate and the correlation coefficient was-0.957.Using principal component analysis and cluster analysis comprehensive evaluation.The order of grafting compatibility of different stock-scion combinations from strong to weak was : ‘Suxin 1’ / ‘Shanhe1’ > ‘Red Earth’ / ‘Shanhe 1’ > ‘Hutai 8’ / ‘Shanhe 1’ > ‘Suxin 1’ / ‘5BB’ > ‘Red Earth’ / ‘5BB’ > ‘Suxin1’ / ‘1103P’ > ‘Hutai 8’ / ‘5BB’ > ‘Hutai 8’ / ‘1103P’ > ‘Red Earth’ / ‘1103P’. |
