Aquilaria sinensis(Lour.)Gilg,a plant in the family Aquilaria family,is a very valuable medicinal and spice product formed after being harmed by the outside world.White wood fragrance is mainly distributed in South China and Southwest China.Since the 70s of the 20th century,wild white wood incense has been predatorily harvested,and wild forests have been greatly damaged,leaving only scattered plants,resulting in a shortage of agarwood.This has led to raw materials having to rely on imports,which are expensive and have a large market gap.In this study,the artificial pure forest of Baimuxiang in Chengmai State-owned Forest Farm in Hainan Province was taken as the research object,and the inducers and pathogenic fungi of Baimuxiang incense were first screened to screen out the compound fungi with the best effect.After that,the suspension cells of Baimuxiang were cultured,and they were induced with a complex fungus,and finally the effect of Baimuxiang incense inducer on trees and suspension cells was verified,in order to obtain a safe,fast and environmentally pollution-free inducer of Baimuxiang incense.In order to meet the needs of the rapidly developing agarwood planting industry and related industries in China,the efficient technology of inducing incense knotting with white wood incense was explored,and the expansion of the fungus library of incense induced by white wood incense was explored.The main findings are as follows:(1)Screening of pathogenic fungi and preparation of inducers in the leaves of C.alba.After injection of different biological agents or chemical agents,the pectinase saturated water solution was selected as the inducer of incense formation.Two fungi,(Pestalotiopsis sp.),and(Phyllosticta sp.),were isolated from the leaves of A.officinalis by tissue separation.The compound can effectively induce the fragrance of C.officinalis.(2)Culture of suspended cells of Radix alba.The best disinfection method for the leaves of C.mongolica was:rinse with water for 2h,then transfer the leaves to the aseptic operation table,sterilize with 75%ethanol for 30s,rinse with aseptic water for 2 times for 30s each time,transfer to 0.1%liters of mercury for 8 minutes,rinse with aseptic water for 5 times for 30s each time.This treatment has the lowest pollution rate of 6.67%.Using MS and WPM medium as the base medium,different hormones were added to perform concentration gradient test,and the optimal medium formula was MS+NAA0.5mg/L+6-BA1.0mg/L+KT0.6mg/L to induce callus formation in the leaves of A.japonicum.The suspension cell system of Xylem sinensis was established successfully.(3)Analysis of the effect of incense formation after induction.The yield of alcohol-soluble extract was 18.3%and 6.5%,respectively,in line with the Chinese Pharmacopoeia standard of more than 10%alcohol-soluble extract.All the alcohol-soluble extracts showed fuchsia,which indicated that the two extracts were in line with the standards of Chinese Pharmacopoeia.The results of optical microscopy showed that the yellow brown agarwood was found inside the cells of C.sinensis.A large number of chromones,sesquiterpenoids and aromatic compounds were detected in all treated ether extracts.These results indicated that the compound bacteriological agent could effectively induce the aromatic formation of C.sinensis.
