Abstract/Summary:
Based on RAD seq analysis results,the study used 100 castor line populations as materials to develop SNP markers,and conducted genome-wide association analysis(GWAS)on main agronomic traits in castor through gene data and phenotypic data,to explore the genes associated with main agronomic traits in castor,in order to provide reference for analyzing the molecular mechanism of the formation of traits,improving main agronomic traits of castor and breeding high-yield varieties of castor.The main results are as follows:1.In this study,991582 high-quality SNPs were filtered and screened using RAD-seq sequencing,and we conducted genome-wide association analysis(GWAS)combined with 10 main agronomic traits of castor including plant height,main stem height,leaf color,wax powder,stem color,stem thickness,leaf vein color,ratio of male to female flowers,main panicle length,and fruit thorns.Finally,a total of 1613 loci significantly associated with the above traits were detected.2.By annotating the gene function of the trait-associated locus region,a total of 959 trait-associated candidate genes were obtained in this study,of which 157 plant height,87 main stem height,288 leaf color,178 wax powder,19 stem color,130 stem thickness,35 leaf vein color,21 female to male flower ratio,25 main spike length,and 19 fruit prickles.3.This study conducted KEGG enrichment analysis on 959 candidate genes,screened out pathways associated with traits from high scoring pathways,identified trait associated genes enriched in them,and performed Quantitative Real-time PCR(q RT-PCR)on trait associated genes.Four pairs of genes related to castor stem diameter and thorn traits were screened: evm.TU.Chr3.820,evm.TU.Chr5.2448,evm.TU.Chr9.523,evm.TU.Chr7.1310.Among them,evm.TU.Chr3.820 and evm.TU.Chr5.2448 are coffee acyl coenzyme A O-methyltransferase(CCo AOMT)involved in lignin synthesis.evm.TU.Chr9.523 is an important peroxidase in the front-end of lignin production.evm.TU.Chr7.1310 is β-Glucosidase,which participates in the process of plant lignification in higher plants.4.In this study,castor was grouped according to the traits of stem thickness and the presence or absence of fruit thorns,and the lignin content was determined.The results showed that the lignin content of castor with thick stems was significantly higher than that of castor with thin stems in the thick stem group;the lignin content of castor with fruit thorns was significantly higher than that of castor without fruit thorns in the thorn group.
