Effects Of Different Immune Enhancers And Oil Emulsion Adjuvants On The Immune Effect Of Foot-and-mouth Disease Vaccine

In order to get rid of the shackles of foreign adjuvant manufacturers,promote the development and production of adjuvant vaccine in China,and improve the quality of foot-and-mouth disease vaccine,in this study,O/RV-1 strain antigen and A/RV-2 strain antigen were added SR01,MR,TH-2,Z-3 immune enhancer respectively,and emulsified with 201 adjuvant to prepare A vaccine containing 20μg 146S per dose.O/RV-1 strain antigen and A/RV-2 strain antigen were emulsified with 201 adjuvant,SW adjuvant,AJW adjuvant,ADJ adjuvant,18C adjuvant and WR adjuvant to prepare vaccines containing20μg and 6μg 146S per dose,respectively.The finished vaccines were tested for physical properties,146S,endotoxin and total protein.The qualified vaccines were those with SR01,MR and TH-2 immune enhancers,and the qualified adjuvant vaccines were 201,SW,ADJ and WR adjuvant vaccines.The disqualified vaccines were the Z-3 immunobooster vaccines,and the disqualified adjuvant vaccines were AJW and 18C adjuvant vaccines.A total of 120 pigs(FMD ELISA antibody titer<1:8)were divided into two groups:group A and group B.Group A was immunized with different immune enhancers and 201 adjuvants.Group B was immunized with different oil emulsion adjuvants and divided into B1and B2groups.Group B1was immunized with 146S 20μg/head vaccine.Animals in group B2were immunized with 146S 6μg/head vaccine.Serum antibodies were detected by ELISA,and the titers of serum O and A antibodies against FMD were compared between group A and group B.The results showed that the vaccines with the highest antibody titer were SW adjuvant vaccine and SR01 immunobooster vaccine.The ELISA antibody titer of FMD in 50 healthy pigs(was determined by ELISA.1:8)were divided into 10 groups((1)to(10))with 5 pigs in each group.Groups(1)to(4)were immunized with SR01 immunobooster vaccine,a total of 20 pigs,2m L/head.(5)to(8)groups were immunized with vaccine without immune-enhancer,20 pigs in total,2m L/head.(9)(10)group was set as the control group(not immunized)with 10 pigs.After 28 days of immunization,the(1)(5)(9)group was injected with the O/Mya98/XJ/2011 strain at the dose of 5000 ID50.(3)(7)(10)group was injected with A dose of 5000 ID50of foot-and-mouth disease strain A/QH/2013;(2)(6)Group was injected with the O/Mya98/XJ/2011 strain of FOOT-and-mouth disease at the dose of 10000 ID50;(4)(8)Group A/QH/2013 strain of FOOT-and-mouth disease was injected with 10000 ID50.The method of challenge was intramuscular injection behind the ear root for 10 days.The results showed that only the unimmunized pigs developed the disease,and none of the immunized pigs showed any clinical symptoms of foot-and-mouth disease,and PCR results showed that no amplification bands appeared in the immunized group.
