The purpose of this thesis is to explore the effect and regulation mechanism of mulberry leaf flavonoids supplementation on oxidative stress status in horses exercise.12Mongolian horses were selected to detect the blood levels of horses before exercise,at different exercise distances and at different time points after exercise.The changes of routine,blood biochemical indexes and blood antioxidant indexes were used to clarify the level of oxidative stress in horses during exercise,and then 12 horses were equally divided into three groups,4 horses per group:the control group and the low-concentration mulberry leaf flavonoid addition group(5g/d/horse).And the high concentration mulberry leaf flavonoids group(10g/d/horse),a 3×3 latin square test was carried out,each feeding period was 20 days,and the exercise test was carried out on 1 day of the test period,During and after exercise,blood routine,blood biochemical indexes,blood antioxidant indexes,muscle fiber structure,muscle antioxidant indexes and gene expression levels were explored to explore its antioxidant mechanism,so that mulberry leaf flavonoids can be used as natural antioxidants to alleviate the oxidative stress of horses during exercise,provide a scientific basis.The results showed that:1.By exploring the changes of blood index levels in horses before exercise,during exercise(5km,10km,15km,20km)and 1h,2h,4h,and 6h after exercise,it was found that the number of white blood cells gradually increased with the increase of exercise distance.,at 20km,it was significantly higher than that before exercise(P<0.05),the highest value reached 14.33×10~9/L at 2h after exercise,and the value decreased to12.70×10~9/L at 6h after exercise,but it was still significantly higher than that before exercise(P<0.05).;The number of lymphocytes and the percentage of lymphocytes first increased and then decreased,and were significantly lower after exercise than during exercise(P<0.05).Hemoglobin and hematocrit increased significantly during exercise(P<0.05),and decreased significantly after exercise and returned to pre-exercise levels;triglyceride and creatinine increased first and then decreased(P<0.05).During the process,it was significantly higher than that before and after exercise;creatine kinase first increased and then decreased(P<0.05),which reached the highest at 2h after exercise,and there was no significant difference between 6h after exercise and before exercise(P>0.05);Antioxidant capacity,superoxide dismutase and glutathione peroxidase gradually decreased from pre-exercise to 20km,gradually increased from 1h to 6h after exercise,and returned to the pre-exercise level;catalase increased from pre-exercise The value gradually decreased until 20km of exercise,significantly lower than before exercise for 20km(P<0.05),and gradually increased after exercise,and was significantly higher than that of exercise 20km at 6h after exercise(P<0.05).With the increase of exercise distance,it gradually increased,reaching the highest at 20km,and gradually decreased after exercise.The above results indicate that a certain degree of oxidative stress occurs during the exercise of Mongolian horses.2.By adding mulberry leaf flavonoids to the diet to explore the changes in blood index levels of horses at different exercise distances and after exercise,it was found that after adding mulberry leaf flavonoids to the diet,the average heart rate and average speed of the horses in the high concentration group were higher when they exercised for 5 km.Compared with other groups;when exercising for 20 km,the average heart rate of the low concentration group was higher than that of the other two groups;the number of white blood cells,monocytes,calcium,and aspartate aminotransferase in the high concentration group were lower than those in the control group(P<0.05),lymphatic The number of cells and platelets in the control group were significantly higher than those in the high concentration group before exercise and at 5km after exercise(P<0.05).The number of neutrophils and total protein in the control group were significantly higher than those in the high concentration group before and 4 hours after exercise.(P<0.05),the number of red blood cells in the control group was significantly higher than that in the high-concentration group before exercise,1h and 4h after exercise(P<0.05),and the hemoglobin in the control group was significantly higher than that in the high-concentration group at 20km and 1h and 4h after exercise group(P<0.05),the hematocrit in the control group was significantly higher than that in the high-concentration group at 5km of exercise(P<0.05),and the triglyceride and total cholesterol in the control group were significantly higher than those in the high-concentration group at 20km and 1h after exercise(P<0.05),the control group was significantly higher than the high-concentration group(P<0.05)when urea exercised for20 km,and the creatinine in the control group was significantly higher than that of the high-concentration group(P<0.05)at 4 hours after exercise(P<0.05).Before exercise,20km and 4h after exercise,the control group was significantly higher than the low-concentration group and high-concentration group(P<0.05).The above results show that the addition of mulberry leaf flavonoids in the diet can improve the antioxidant level of Mongolian horses to a certain extent,and the addition of high concentrations of mulberry leaf flavonoids has a better effect on alleviating oxidative stress in Mongolian horses.3.By exploring the changes of muscle index levels in horses after adding mulberry leaf flavonoids after exercise and 24 hours after exercise,it was found that compared with the control group,the high concentration group had higher levels of total antioxidant capacity,superoxide dismutase,catalase,glutathione and glutathione.The expression levels of peptide peroxidase,Trx R1,SOD1,SOD2,GPX1,GPX3,and Nrf2 were all increased,and the levels of malondialdehyde and muscle fiber damage were decreased.Enzyme gene expression,thereby promoting the activity of antioxidant enzymes,improving the antioxidant performance of Mongolian horses,and alleviating muscle fiber damage caused by oxidative stress in Mongolian horses.To sum up,the Mongolian horse produces oxidative stress in the process of movement,mulberry leaf flavonoid is added in the diet can significantly improve Trx R1,SOD1,SOD2,GPX3 antioxidant gene expression quantity,increase the superoxide dismutase,catalase and glutathione peroxidase content,enhance the body’s total antioxidant capacity,alleviate sports oxidative stress damage,Among them,the high concentration of mulberry leaf flavonoids added 10g/d/horse group had better effect. |
