| Prolactin(PRL)has the function of regulating the hair follicle growth cycle,it can directly or indirectly affect the activity of the hair follicle,promote hair growth and speed up the development of the hair follicle to the degeneration stage.In this study,the hair follicles of Inner Mongolia cashmere goat were taken as the research object.Prolactin was added to the hair follicle culture in vitro to observe the growth of the hair follicles and analyze their morphological structure.The hair follicle transcriptomes of the prolactin group and the control group were sequenced by GO analysis and Pathway analysis,and the Differentially expressed genes(DEGs)related to hair follicle growth and development were screened as candidate genes.The expression of these candidate genes at m RNA level was analyzed by real-time fluorescence quantitative PCR technology,so as to explore the influence mechanism of prolactin on hair follicle growth and development.The main results are as follows:1.Differentially expressed genes of the control group(K group)and prolactin group(PRL group)were screened by analyzing the transcriptome sequencing data.A total of2488 differential expressed genes were screened in the primary hair follicle group(PF-K vs PF-PRL),and 1435 genes were up-regulated and 1053 genes were down-regulated in the prolactin group compared with the control group.A total of 1722 differentially expressed genes were screened in the secondary hair follicle group(SF-K vs SF-PRL),compared with the control group,470 genes were up-regulated and 1252 genes were down-regulated in the prolactin group,including 41 DEGs related to hair follicle development.29 in the primary hair follicle group and 12 in the secondary hair follicle group,which were enriched in MAPK,Wnt,Hedgehog,Notch,JAK-STAT,NF-KB and other follicle-related pathways.2.When the prolactin concentration was 10 ng/m L,the average hair follicle growth length was significantly higher than that of other groups(0/5/50/100 ng/m L)(P<0.01),10 ng/m L prolactin had the best promoting effect on hair follicle growth.The stem and root sheath of primary/secondary hair follicles in PRL group and K group were elongated simultaneously,and bending appeared in different degrees with the increase of culture time.Compared with K group,the length of primary/secondary hair follicles in PRL group had no significant difference from 0 to 1d(P>0.05),but the length of primary/secondary hair follicles in PRL group increased significantly from 2 to 5d(P<0.01).3.Qrt-pcr was used to detect the expression levels of differentially expressed genes FGF9,IL8,SOCS4,DLL4 and SFRP1,CNTFR,IL6,SMAD9 in the control group and prolactin group,and the results showed that prolactin significantly increased the expression of CNTFR(P<0.05),significantly increased the expression levels of SFRP1,IL6 and SMAD9(P<0.01),and significantly decreased the expression levels of FGF9,IL8 and DLL4(P<0.01)had no significant effect on SOCS4 expression(P>0.05).In addition to SOCS4,several other genes are involved in the regulation of hair follicle growth and development. |
