Abstract/Summary:
In order to understand the genetic rule of faba bean starch content and discover the QTL loci related to faba bean starch content,this study used the established F2population as the experimental material to carry out genetic analysis of faba bean starch content.Using F2populations and natural populations,combined with SSR marker,linkage analysis and association analysis were carried out on the starch content of faba bean,in order to provide support for the selection and MAS breeding of starch-specific faba bean varieties,thereby speeding up the breeding process and promoting the quality of faba bean breeding.The main findings are as follows:1.Taking Afghanistan’496’(high starch content material)and China Zhejiang’Zhongzi’(low starch content material)as parents,F1and F2hybrid populations were prepared,and the starch content of faba bean was genetically analyzed by 4 generations.The results showed that the inheritance of grain starch content in faba bean conformed to the MX2-EAD-AD model(2 pairs of additive-dominant major genes+additive-dominant polygenic model).The additive effect value and dominant effect value of the two pairs of main genes are equal,both are 1.686,the additive effect value and dominant effect value of polygene are 1.0705and-4.1252,respectively,the genetic variance of the main gene is 3.9225,and the heritability is 52.56%,heritability is small.2.A genetic map of faba bean containing 9 linkage groups and 124 SSR markers was constructed.The map coverage length was 1321.43 c M,the length of each linkage group was between 44.86 and 295.46 c M,the average map distance was10.66 c M,and the number of linkage group markers was 5~22.Using ICIM to analyze the starch content of faba bean,five QTL loci(q SC1,q SC2,q SC3,q SC4,q SC5)related to starch content were detected,which were located in the LG02,LG06and LG07 linkage groups,respectively.The interpretation rate of phenotypic variation They were 3.7%,3.39%,3.4%,0.44%,and 2.6%,respectively,explaining a total of13.54%of the phenotypic variation.3.Using 133 pairs of SSR markers with polymorphism to analyze the population genetic structure of 333 faba bean germplasm resources,it was found that the natural population was divided into two subgroups.The association analysis of starch content and markers in 2019 and 2020 was performed using GLM and MLM models,and a total of 14 markers significantly(P<0.05)associated with starch content were detected.Among them,the SSR-10927 marker was detected to be significantly associated with starch content in both two years and both models.Three markers(V2188,V2228,V1133)were detected in both models at the same time and were significantly associated with starch content in 2020(P<0.01),with explanation rates ranging from 4.31%to 7.75%(GLM)and 3.57%to 6.19%,respectively.(MLM).
