| Object:In this study, cotton stalk was used as raw material,by adding different proportions of molasses,Lactobacillus plantarum, quantitative salt and urea for fermentation, the purpose of this study was to investigate the fermentation quality, nutritional quality, microflora and rumen degradation rate of fermented feed with different ratios of molasses and Lactobacillus plantarum to screen the best combination treatment, provide scientific guidance for the fermentation of cotton stalk and molasses with lactic acid bacteria. Methods:The test adopts bagged micro-storage technology, set different molasses addition levels(0%,2%,3%,4%,5%,6%),different molasses(4%,5%)and Lactobacillus plantarum addition levels(0%,0.1%,0.2%,0.3%),dynamic detection of its nutrients, fermentation indicators, determination of aerobic stability, microflora and nutrient degradation rates of fermented products treated with a combination of molasses and Lactobacillus plantarum by nylon bag method.Results:(1)The addition of molasses had a significant effect on the micro-storage of cotton stalk, the WSC content increased, the higher CP content was retained, the LA content increased, and the p H decreased. At the same time, compared with the control group, the AN/TN value,FG,ADF and NDF were reduced, and the fermentation quality was improved. As the amount of molasses added increases, the abundance of bacteria of the phylum Firmicutes,Lactobacillus and Levilactobacillus gradually increased, the abundance of fungus of the phylum Ascomycota,Alternaria and Cladosporium decreased gradually, increased abundance of Wickham yeast, effectively inhibiting the growth of spoilage microorganisms.Considering the fermentation quality and cost saving,it is more appropriate to add 5%molasses.(2)Compared with the pure 5% molasses group, the content of CP,LA and WSC in the group added with 5% molasses and 0.2% Lactobacillus plantarum increased 2.56%,5.67%,5.49%,p H,AN/TN value,ADF,NDF content decreased by 0.48%,1.30%,5.01%,2.11%.With the addition of molasses and Lactobacillus plantarum increased, the abundance of bacteria of the phylum proteobacteria decreased, increased abundance of Lactobacillus,Pantoea,Leuconostoc;the abundance of fungus of the phylum basidiomycota increased, decreased abundance of Alternaria,Cladosporium and Botrytis.Considering the fermentation quality and cost saving,it is suitable to add 5% molasses and 0.2% Lactobacillus plantarum.(3)The 72 h rumen degradation rate and effective degradation rate of DM,NDF and ADF in5% molasses,0.2% Lactobacillus plantarum group were the highest, the membership function value of the effective degradation rate of nutrient components ranked first, and it was known to be the best fermentation combination. Conclusion: Adding molasses, molasses combined with Lactobacillus plantarum can improve the quality of micro-storage;When the addition amount of molasses and Lactobacillus plantarum was 5% and 0.2%,the fermentation quality of cotton stalk was significantly improved,it can change the microflora and increase the proportion of lactic acid bacteria in the fermentation system, the rumen degradation rate and effective degradation rate of nutrients DM,NDF and ADF were the highest among the fermentation products of each group. |
