Rosa multiflora and rosa belong to the Rosa in Rosaceae,Modern Rose is their hybrid,one of the four world’s major cut flowers.It is popular for the beautiful shape and color,However,Powdery Mildew seriously reduce the yield and ornamental traits of cut flowers.Through the application of chemical agents to control powdery mildew is the most important way,which seriously destruct the environment,while the cost is too high,Therefore,by breeding,we need to introduct the better genetic resources to broaden the genetic basis and make up for the lack of resistance characteristics of Rosa,In this study,we studied the gene expression patterns of the resistant and susceptible moltiflora.And constructed the VIGS and over expression vectors that would be involved in the gene function verification,In order to achieve better resistance in Rose to powdery mildew.The results are as follows:(1)The resistant and susceptible materials,a total of 20380 differential genes at 0h,of which 10196 were upregulated and 10184 were downregulated,In susceptible materials,a total of 6785 different genes were found at 24h;4269 were upregulated,2516 were downregulated at 24h;A total of 5971 differential genes,3603 were upregulated,2368 were downregulated at 96 h,In resistant materials,7516 different expressed unigenes,5036 were upregulated and 2480 were downregulated at 24h;12464 different expressed unigenes,8836 were upregulated and 3628 were downregulated at 96h;The enriched analysis of KEGG showed that the differential genes were enriched with a large number of metabolic pathway in resistant and susceptible materials,the flavonoid pathway and plant pathogen interaction are mainly enriched in resistant material,The plant hormone signal pathways is enriched in susceptible material,The gingerol pathway may only provide intermediate material for other metabolic pathways in disease resistance.(2)We take GADPH as a internal reference gene to apply quantitative real-time PCR.Found that the expression patterns of 12 candidate genes are different between resistant and susceptible materials.the flavonoid pathway、 salicylic acid pathway and jasmonic acid pathway are more obvious in resistance;Lignin synthesis pathway is more obvious in susceptible materials The results showed the expressing patterns of 12 genes were different in the resistant and sensitive plants,which may take important roles in disease resistance.The resistant material may respond faster to powdery mildew.(3)The correlation analysis between the transcriptome and proteome showed that the correlation degree was very low,at the 24 h,correlation coefficient was 0.1729 and 0.2889 respectively in resistant and susceptible plant.The correlation coefficient of was respectively 0.3364 and 0.1544 at 96 h in resistant and susceptible plant,which indicated that the regulation of post-transcriptional was significant,At the same time,6 and15 proteins were identified in resistant and susceptible materials,which were related to flavonoid pathway,plant and pathogen pathway,plant hormone pathway and Stilbenoid,diarylheptanoid and gingerol biosynthesis,The protein AOS and MPK4 were identified in the susceptible material.(4)Five VIGS vectors were successfully constructed.At the same time,the two Overexpression vectors were constructed.
