| The potato industry in Enshi occupies an important position in the agricultural production of Hubei Province,but the virus diseases have seriously hindered the development of the local potato industry.To investigate the prevalent potato virus diseases in Enshi,442 potato tubers reserved by farmers were collected as materials from 118 sampling points at different altitudes and regions as materials.The main potato virus diseases in Enshi were analyzed by using the enzyme-linked immunosorbent assay and Next-generation sequencing technology,and the diversity of related virus strains was further analyzed by reverse transcription polymerase chain reaction technology,which provided the basis for potato introduction,breeding and virus disease control in Enshi.The main research results are as follows:1.The incidences of PVS and PVM were 53.5%and 23.0%respectively,in tuber samples collected in Enshi,and the two viruses were the main epidemic potato viruses.Meanwhile the incidences of PVA,PVX,PLRV and PVY were all lower than10.0%.In addition,PVH and PMTV were detected in few samples by sequencing and RT-PCR.The frequency of PVY virus was higher in mountain below 700m than that in higher mountain area,PVX was the most frequent virus in mountain 700-1000m high,and PVS and PVM virus were more common in mountain above 1000m.In addition,the detection rates of PVY,PVX,PLRV and PVA viruses were significantly lower than those below 1200 m,while the frequency of PVM and PVS was higher above 1200 m.By region,the samples from Badong and Jianshi were infected with PVS and PVM most seriously,while PVX and PVA were more prevalent in Hefeng2.The strain diversities of PVS and PVY was analyzed by RT-PCR.The results showed that 59.3%PVSOstrain and 29.2%PVSAstrain were detected in 113 PVS positive samples,and a small amount of mixed infection of the two PVS strains was detected.In 23 PVY positive samples,60.8%PVYNTNstrain,21.7%PVYN:Ostrain and 8.7%PVYNstrain were detected.These results indicated that PVSOand PVYNTNwere the main epidemic strains of PVS and PVY in Enshi,respectively.3.The PVM and PVA genotyping methods based on RT-PCR were established to investigate the strain diversity in samples collected in Enshi.The results showed that 70.0%of the 70 PVM positive samples were PVMOstrains,and a small number of PVMK,PVMTand PVMD1strains were detected,but they all were mixed infection with PVMOstrains.Among the 36 PVA positive samples,mixed infection of PVAAand PVAWstrains were the dominant,followed by mixed infection of PVAA,PVAWand PVATstrains,accounting for 61.1%and 33.3%,respectively. |
