Abstract/Summary:
With the development of technologies,more and more studies on rice transcriptome were conducted using high-throughput sequencing technologies in recent years.The short read sequencing technology limited its applications in studies to identify alternative splicing and fusion genes.Full length transcripts can be obtained using single-molecule real-time sequencing technology,avoiding errors caused by transcripts assembly.In this study,transcripts of ZS97,MH63,F1 hybrid and F1 reciprocal cross were sequenced by single-molecule full-length transcript sequencing technology.The main research contents are as follows:(1)Transcriptome sequencing data of four rice samples were processed,and 3,834,218Reads of Insert were obtained.A total of 1,730,745 full-length transcripts were identified,and 397,478 high accuracy transcripts were obtained after clustering and correction.(2)Based on the high accuracy transcripts,we analyzed alternative splicing events in all sequencing data.We found that about 60%of the transcripts of ZS97 and MH63undergone alternative splicing.The percentages(54%)of alternative splicing of F1 hybrid and F1 reciprocal cross were lower than that of the parents.Intron retention was the most common splicing event for all four rice cultivars.(3)Potential Hybrid/Fusion Transcripts were identified in the sequencing data of two F1hybrid.A total of 1,037 and 998 Hybrid/Fusion Transcripts were identified for the F1hybrid and the F1reciprocal cross respectively.
