As an important cash crop,castor beans is widely distributed all over the world.Castor oil from castor seeds is of great economic value to industry.Castor beans have good adaptability to the surrounding environment,and one of the biggest management problems faced by large areas of castor beans is disease.Castor gray mold is a highly contagious and destructive plant disease.Botrytis cinerea infects castor flowers more easily.When the disease was serious,the corrupt flower directly led to a large number of reduction of output.WRKY transcription factor family is a typical regulatory factor regulating plant environmental adaptability.WRKY33 is an important immune regulatory factor in plants and plays a key role in the study of plant resistance to B.cinerea.In order to analyze the relationship between WRKY of castor bean and its resistance,all members of the WRKY family of castor bean were extracted from the whole genome data of castor bean,and the potential function of WRKY33 homologous protein in castor bean against gray mold was predicted.The conclusions of this study are as follows:RcWRKY46 and RcWRKY51 homologous genes of WRKY33 in Castor bean were screened by bioinformatics method.The expression analysis of castor bean after inoculation by B.cinerea,Several WRKY genes were up-regulated in castor bean,among them,RcWRKY46 and RcWRKY51 were significantly induced by B.cinerea.The representative genes RBOHD and ORA59 in the resistance pathway were also significantly upregulated.RcWRKY46 and RcWRKY51 genes and their promoters were cloned.In order to verify the RcWRKY46 and RcWRKY51 promoter activity,GUS recombinant vectors were constructed and transient expression in castor bean showed that the promoter was active.Genetic transformation in castor bean using CRISPR system,The false positive plants were analyzed and discussed.In order to further verify the biological functions of RcWRKY46 and RcWRKY51.Overexpression vectors of RcWRKY46 and RcWRKY51 were constructed to transgene Arabidopsis wrky33.The overexpressed lines of RcWRKY46 and RcWRKY51 were obtained.Infection experiments on the identified overexpressed lines showed that compared with the high sensitivity of wrky33 to B.cinerea,the overexpressed lines recovered certain resistance.It is of great value to cultivate superior varieties for disease resistance of castor bean.This study provided a preliminary basis and certain reference for the further study of the biological functions of WRKY gene family in castor bean and the resistance of WRKY33 homologous protein in castor bean.
